| Coleus Forskolin Briq is a herbal grown in YuNan, which has the effects of "xuanfei", anti-pyrexia and "jiangni"anti-asthma. This experiment is desired to confirm the anti-asthma action of Coleus Forskolin Briq, and to carry out a preliminary study with its anti-asthma mechanism. This experiment investigates the anti-asthma action of Coleus Forskolin Briq to bronchial asthma using asthmatic model (asthmogenic method by histamine nebulization, lung overflowing model of guinea pig, et al) and in vitro model (isolated trachea strips of guinea pig); studies the anti-inflammation and antibiosis action of Coleus Forskolin Briq (sponge granuloma test of rat and in vitro antibiosis test); investigates the " fuzhengguben" action of Coleus Forskolin Briq by its effect to immunological function, hypoxia and normal atmosphere pressure tolerant capacity and burden swimming time of mice; and preliminarily approachs the anti-asthma mechanism of Coleus Forskolin Briq, which focuses on anti-oxidation and cytokine causing asthma, using chronic bronchial asthmatic model of rat sensitized with oval protein(intraperitoneal oval protein saline 1 mL which contain 7 xl09 inactivated B.pertussis on normal male rats. From the fifteenth day, inhaled 10 g/L oval protein aerosol on the rats for 20 min, exciting serially for 1 w. The last time, killed the rats 1 h after inhaling, took serum and lung douche liquor to assay the content of SOD, MDA and IL-4).Asthmogenic method by nebulization shows that Coleus Forskolin Briq high dosage group (12.8 g raw herbal-kg"1) and middle dosage group (6.4 g raw herbal-g"1) can obviously prolong the asthmatic time induced by histamine (p<0.01), and this action correlates to the dosage; Coleus Forskolin Briq can obviously decrease the range of guinea pigs' lung overflowing curve induced by histamine; Coleus Forskolin Briq can obviously inhibited the contractions induced by histamine; the result shows that Coleus Forskolin Briq play a significant role in anti-asthma.Coleus Forskolin Briq effect little on rat sponge granuloma, which points out that Coleus Forskolin Briq has no effect on inflammation caused by foreign material. However,compared to control group, Coleus Forskolin Briq high and middle dosage group can increase animals' spleen index and thymus index, which imply that Coleus Forskolin Briq can prove immunity; in vitro antibiosis test shows that Coleus Forskolin Briq can resist Achromobacter xylosoxidans, staphylococcus aureus and aeruginosa Pseudomonas, and MIC are 0.1 g-mL'1, 0.1 g-mL'1, 0.5 g-mL ~].Coleus Forskolin Briq can increase spleen index and thymus index of rat, increase lymphocyte conversion induced by PHA, and enhance the conversion rate.; carbon clearing up test shows that Coleus Forskolin Briq can enhance the index and activity of phagocytosis of mice, which imply that it play a role in improving cellular immunity , monocyte phagocytosis function and non-specific immunity of mice; compared to control group, the survival time under the condition of hypoxia and normal atmosphere pressure of mice in high dosage elongates obviously (p<0.01), and so is the burden swimming time (p<0.01). the result shows that Coleus Forskolin Briq has the action of " fuzhengguben " ?Using Coleus Forskolin Briq intervene the chronic bronchial asthmatic model of rat sensitized with oval protein, we investigate that the activity of SOD in serum and lung douche liquor increased, MDA in serum decreased, which implies that Coleus Forskolin Briq can improve the anti-oxidation function of body, remove oxygen-derived free radidicals, and inhibit lipid peroxidation; IL-4 in rat serum and lung douche liquor of Coleus Forskolin Briq group is fewer than that of control group, which shows that Coleus Forskolin Briq can inhibit IL-4, relieve inflammation in rats' airway, and decrease hypersensitivity of airway.In brief, Coleus Forskolin Briq has good action of anti-asthma both in asthmaticmodel and in vitro model, has certain action of antibiosis, enhance immunity," fuzhengguben " ,...
