ObjectiveFunction constipation is a very common disorder or disease of digestive tract, Great interests have been focused on the diagnosis and therapy of this disease. From the functionl changes to substantiality lesions of constipation; from gastrointestinal hormones,neural transmitters,enteric nervous system to the study of cajal cells; from laxative abuse to the Practice Guidelines for Chronic Constipation of China published; from conservative therapy to operative intervention for the treatment of constipation, the level of diagnosis and treatment and basic research for constipation in China is guadually increasing. But so far there are few reports about the etiopathogenisis and progression of constipation from the view about the changes of water absorption and mucus secretion in large intestine. There is also no systemicand relative study about aquaporins (AQPs) in large intestine.so we study the expression and distribution of aquaporin 3 and aquaporin 9(AQP3 and AQP9) in mucous membrane of colon, to study the relationship between them and constipation, and provide new target point of clinical diagnosis and treatment for patients with functional constipation.MethodFirst of all,to selected 45 patient who sufferered constipation and 32 health people,immunohistochemistry technique was used to detect the distribution of AQP3 and AQP9 in mucous membrane of colon. Then, Western blotting relatively semi-quantitative technique was used to detect the level of AQP3 ,AQP9 andβ-actin was used as interior reference, the ratio of AQP3,AQP9 andβ-actin is their relative amount, statistics analysis was used to the ratio of gray scale. to use SPSS11.0 statistical package goes to statistics analysis, the data was Gaussian distribution,T test of two independence samples, test criterion a =0.05Result1 .There was a positive expression of AQP3 and AQP9 in the left and right colonic mucosa of patients with functional constipation (testing group) and subjects without constipation (control group).AQP3 distributed mainly in the basement membrane of epithelial cell of mucous membrane of colon;AQP9 distributed mainly in the goblet cell in mucous membrane cavosurface.2. The average ratios of gray scale of AQP3 in left colon and right colon of control group were 0.701±0.013 and 0.798±0.012 respectively, which had statistical significance. That of testing group were 0.697±0.010 and 0.905±0.017 respectively, which had statistical significance. The ratios of gray scale of AQP3 in the right colon between testing group and control group had not statistical significance. That in the left colon between testing group and control group had statistical significance.3 . The average ratios of gray scale of AQP9 in left colon and right colon of control group were 0.732±0.012 and 0.543±0.010 respectively, which had statistical significance. That of testing group were 0.544±0.015 and 0.545±0.013 respectively, which had not statistical significance. The ratios of gray scale of AQP9 in the right colon between testing group and control group had statistical significance. That in the left colon between testing group and control group had not statistical significance.Conclusion1. There were expression of AQP3 and AQP9 in the left and right colonic mucosa of the man.AQP3 distributed mainly in the basement membrane of epithelial cell of the right colon mucous membrane;AQP9 distributed mainly in the goblet cell of the left colon mucous membrane. AQP3 has something to do with the absorption of water, but AQP9 have something to do with the secretion of mucus.2. The ratios of gray scale of AQP3 in the left colon are lower than the right colon in control group, the test group are same to the control group, the ratios of gray scale of AQP3 of the test group increases in the right colon, but that is not change in the left colon, to hint the absorption of water are located in the right colon.3. The ratios of gray scale of AQP9 in the left colon are higher than the right colon in control group, but that are not difference in the test group, the ratios of gray scale of AQP9 of the test group are not change in the right colon, but AQP9 decreases in the left colon ,to hint the secretion of mucus are located in the left colon.4. The increase or decrease of the amount of AQP3 and AQP9 may have relationship to the occurrence and development of constipation.
