Objective:To study the effects of the different active-site proportion components of the total alkaloids from leonurus and astragaloside on benign prostatic hyperplasia (BPH) mice and rats models by baseline design compares increase or decrease. To perfect chinese medicine could improve component ratio optimization selection mode and screening the optimal active ratio components to prevent BPH. Subsequent research lay the foundation for the Motherwort total alkaloids-Astragaloside capsule.Methods:screening tests:To study the effects of the different active-site proportion components of the total alkaloids from leonurus and astragaloside10:0、8:2、7:3、6:4、5:5、4:6、3:7、2:8、0:10on BPH model of mice and BPH model of rats by baseline design compares increase or decrease.Using comprehensive weight method evaluate the component ratio of the total alkaloids from leonurus and astragaloside on BPH mice and rats models, screening The optimal active ratio components. Validation experiment:Though the test of model of mice and rats validate screening experimental results.Using comprehensive weight method evaluate the experimental results.The optimal active ratio components of the total alkaloids from leonurus and astragaloside to prevention BPH would be determined.To establish BPH model of mice by subcutaneous injecting testosterone propionate induction method.By subcutaneous injecting testosterone propionate5mg/kg(soluble in soybean oil)in mice every day to copy the BPH mice model, and then last21days. After21days,the mice were put to death,observe the changes of prostate weight and prostate exponent.Screening experiment test the content of DHT and histology under light microscope of prostate.Validation experiment test the content of DH、T、E2and the pathologic changes of prostate and the related organization form.Wistar male rats model of BPH were established by subcutaneous injecting testosterone propionate induction method. Rats were castrated, from the eighth day subcutaneous injected testosterone propionate4mg/kg/d for30days. After30days,the rats were put to death,observe the changes of prostate weight and prostate exponent.Screening experiment test the content of T、E2、PACP in blood serum,and histology under light microscope of prostate. Validation experiment test the content of DHT、T、E2、PACP and the pathologic changes of prostate and the related organization form.Results:The result of screening tests:The rats and mice BPH models were reproduced successfully.The different proportion components of the total alkaloids from leonurus and astragaloside can significantly decreased prostate weight and prostate exponent in mice and rats. Significantly improve the prostate of the rats and mice, Significantly reduce the level of DHT in the mice. Significantly reduce the level of T、PACP and obviously rise the level of E2in blood serum of rats. Using comprehensive weight method evaluate, the optimal active ratio is6:4. The result of Validation experiment:the different proportion components of the total alkaloids from leonurus and astragaloside7:3、6:4、5:5dose group of big^middle and small can reduce significantly the level of T and DHT, obviously rise the level of E2in blood serum of mice, significantly improve the the pathological changes of prostate、testis、thymus gland and spleen of mice.5:5dose group of big and6:4dose group of big、middle can significantly decreased prostate weight and prostate exponent in mice.6:4、5:5dose group of big、middle and small can reduce the level of T、DHT and PACP in the prostate tissue of rats, obviously rise the level of E2in blood serum of rats, significantly improve the the pathological changes of prostate、 epididymis、thymus gland and spleen of rats.5:5dose group of big and6:4dose group of big、middle can significantly decreased prostate weight on rats.Using comprehensive weight method evaluate, the optimal active ratio is6:4of the different proportion components of total alkaloids from leonurus and astragaloside.Conclusion:Screening tests:the optimal prevention BPH proportion components is6:4in the different proportion components of total alkaloids from leonurus and astragaloside by baseline design compares increase or decrease; Validation experiment:As the center6:4of total alkaloids from leonurus and astragaloside,each add a ratio and components.Though validate the BPH the model of mice and rats,the result was consistent with the experiment.Finally,the optimal active ratio components to prevente BPH in total alkaloids from leonurus and astragaloside is6:4.
