Objective: To evaluate the effect of the total alkaloids from leonurus japonicus to benign prostatic hyperplasia (BPH) mice and rats models,and evaluate the effect of the stachydrine hydrochloride to benign prostatic hyperplasia (BPH) mice and model. Beside the measurement of the weight, exponent, one prostate and the determination of the testosterone(T), estradiol(E2), basic fibroblast growth factor(bFGF),epidermal growth factor(EGF),ransforming growth factor beta-1(TGF-β1),insulin-like growth factor-Ⅰ(IGF-Ⅰ) content of the animal models, and the microcosmic change of the mice and rats'prostate observe was also observed.To study the pathogenesis of BPH with four growth factors, clarify the mechanism of the total alkaloids and the stachydrine hydrochloride restraining BPH.Methods:The mice model of BPH was also established by urogenital sinus implantation.Male model mice were divided into six groups: control group, model group, Longbishu group, finasteride group, total alkaloids low dose group, middle dose group and high dose group and the stachydrine hydrochloride low dose group, middle dose group and high dose group and then treated with each group's drug at the same time for 21 days. After 21 days, the mice were put to death, observe the changes of prostate weight, prostate exponent, histology under light microscope of each group, measure the DHT,PACP content in blood serum and detect the expression of bFGF,EGF,IGF-Ⅰ,TGF-β1 in prostate.. Wistar male rats were also divided into six groups: control group, model group, Longbishu group, total alkaloids low dose group, middle dose group and high dose group. Rats were castrated, from the eighth day subcutaneous injected testosterone propionate, oestradiol benzoate (3mg:50ug)for 30 days and treated with each's drug at the same time. After 30 days, the rats were put to death, observe the changes of prostate weight, prostate exponent, histology under light microscope of each group, observe the changes of prostate ultrastructure under electon microscope, measure the T, E2,DHT in prostate or blood serum,and detect the expression of bFGF,EGF,IGF-Ⅰ,TGF-β1 in prostate.Results: The BPH mice and rats models were all successfully copied, The prostate weight,prostate exponent in model mice and rats increased greatly(P<0.01), the content of T,DHT in blood serum or prostate constitution in model mice or rats increased greatly (P<0.01),the expression of bFGF,TGF-β1,EGF,IGF-Ⅰin prostate of model mice or rats increased greatly(P<0.01), the volume of prostate hyperplasied greatly(P<0.01), ultramicrostructure of the prostate had a accrementition change.The total alkaloids and the stachydrine hydrochloride can obviously or significantly decreased the level of DHT,PACP and the expression of EGF,IGF-Ⅰ(P<0.05orP<0.01),and obviously rise the expression of TGF-β1 (P<0.05) in model mice. The total alkaloids can obviously or significantly decreased the level of DHT,T and the expression of bFGF,EGF,IGF-Ⅰ(P<0.05orP<0.01), and obviously rise the expression of TGF-β1 (P<0.05) in model rats. In histology the model animals have obvious anomaly, the total flavonoids can relieve the anomaly and improve the prostate ultrastructure.Conclusions:The total alkaloids from leonurus japonicus has good effect on BPH animal models, it can decreased the model animals prostate exponent, decreased the content of T,DHT in model mice or rats, decreased the expression of bFGF,EGF,IGF-Ⅰand rise the expression of TGF-β1, increased TGF-β1/bFGF which could restrain BPH, improve the prostate ultrastructure,relieve the histologic anomaly of the model animals'prostate constitution. The research suggested that the total alkaloids from leonurus japonicus can also treat BPH.We infer that the mechanisms of the total alkaloids to BPH may be have relationship with growth factors,regulation of the proportion of androgen and estrogen. In a word, total alkaloids from leonurus japonicus can restrain BPH.
