The diosmin is a flavonoid which extracted from Galium verum L. and it has ansignificant effi cacy to inhibit venous thrombosis, enhance venous tone, treatlymphatic system disorders, reduce blending. This study analysis focuses on theradioprotective properties of diosmin, by performing in vitro experiments、in vivoexperiments and using mice liver proteins to find different protein points. Through aseries of experiments, we can obtain the follow research results:(1)On splenic lymphocytes and PC12cells were cultured as a model to study thesplenic lymphocyle proliferation, survival and cellular oxidative damage. Theexperiment prove that diosmin can effectively reduce cell mortality and improve cellviability when at a concentration of80μg/ml before irradiation. While at this dose,diosmin can can effectively reduce cells damage, such as increased superoxidedismutase (SOD),glutathione (GSH-px) activity and reduce intracellularmalondialdehyde (MDA) levels. Through the PC12cells cycle and apoptosisdetermination, we can learn that diosmin can promote cells differentiation and reducethe damage caused by irradiation.(2)By establishing60Co-γ ray irradiation model of continuous gavage14daysdiosmin mice was measured through a series of indicators to determine diosminanti-radiation effect. It was found that diosmin can play a protective role to mice bodyweights, white blood cells, innume organs such the liver and spleen. It also canimprove the activity of SOD, GSH-px, decrease MDA levels, chromosome aberrationand micronucleus rate of mouse tissue. By a series of experiments, diosmin wasenhanced antioxidant activity by improving endogenous antioxidant enzyme activityand can reduce DNAdamage, protects mice from radiation injury.(3) By irradiation experiments in mice liver protective protein2-DE Proteomeanalysis showed that there was33spots increased while35decreased comparing withthe control group. Comparing with the model group, there were82protein spotsincreased while9decreased. It was possible to identify10proteins, which areacetyl-Coenzyme A acyltransferase2, peptidyl-prolyl cis-trans isomerase A,3-hydroxyanthranilate3,4-dioxygenase, Chain B, Structure Of Apo-CalmodulinBound To Unconventional Myosin V, Heterogeneous nuclear ribonucleoproteins A2/B1, Triosephosphate isomerase, Fructose-bisphosphate aldolase B, proteindisulfide-isomerase precursor, hemoglobin subunit beta-1, mitochondrial ATPsynthase, H+transporting F1complex beta subunit.
