Objective:Diosmin can provide a very strong protection for blood vessel,it can inhibit the formation of thrombus and it also has the function of anti-inflammatory and anti-thrombotic.Depends on the previous experiment research of chemical and pharmacodynamic,flavonoids ingredients in Galium Verum L.is the active ingredients for treatment of venous system disease,in addition diosmin is the part of activity indicative of Galium Verum L.Therefore the function of anti-thrombotic about diosmin is the main idea of this subject.Method:The experiment used 70%ethyl alcohol to withdraw the flavonoids ingredients in Galium Verum L.The AB-8 macroporous resin was used to depuration flavonoids ingredients,than separated and refined flavonoids ingredients farther to obtained diosmin.Furthermore,to proceed a pharmacodynamic study for diosmin:grouped 60 Wistar rats in blank set,model set,diosmin low-dose set,diosmin middle-dose set,diosmin high-dose set and aspirin set.Gave each set medicament for 7 days,than the inferior vena cava ligation method was applied to establish lower limb venous thrombosis model.The blood was brought to measured coagulation four indices(PT,APTT,TT,FIB)the day after established the model;Hemorrheology indicators:plasma viscosity,whole blood viscosity,hematocrit,erythrocyte sedimentation rate,red cell assembling index,red cell degeneration index and any other index;Optical microscope and electron microscope was used to observe postcava vascular tissue.Consequence:The experiment used the AB-8 macroporous resin to purification delicately and the yield coefficient of flavonoids ingredients is 24%.Diosmin came from futher separate flavonoids ingredients delicately.Further separation of refined flavonoids part get diosmin,using high performance liquid chromatography(HPLC)method for determining the content of diosmin,then inspect the methodology.This method,use the Shimadzu VP-ODS C18 chromatographic column,mobile phase is methanol-Acetic acid-water(47:3:50);detection wavelength is 275nm,the speed is 1.0 mL·min-1,temperature is 30?,sample quantity is 10?L.The content ofdiosmin is 90.77%.Coagulation four indices:PT:Diosmin set with low-dose,middle-dose and high-dose could remarkable prolong the time about PT,APTT,TT,reduce the content of FIB(P<0.05).Hemorrheology-:Diosmin set with low-dose,middle-dose arid high-dose could reduce whole blood viscosity,plasma viscosity,erythrocyte sedimentation rate,hematocrit,red cell assembling index and red cell degeneration index.Histological observation with optical microscope:Diosmin set with low-dose,middle-dose and high-dose could reduce thrombus in lumen obviously,loosed construction,more completed vein,the endothelial cells complete without a loss,there is no inflammatory cells around the blood vessels.Histological observation with electron microscope:Diosmin set with low-dose,middle-dose and high-dose could keep continuous and complete about endothelial cell,relieved damage situation,had no distinct attrition of endothelium;Had a clear outline of mitochondria,a complete nuclear membrane,a clear ridge structure,had rough endoplasmic reticulum and ribosome.Conclusion:Diosmin could improve the coagulation four indices index and hemorrheology index of thrombus rats.Histological observation also shows that diosmin set with treatment had the impact about the protection of blood vessel.The experiment illustrated that diosmin can confront cruor and phlebothrombosis,it has a good effect for the treatment of thrombotic disorders.It has the same treatment effect on thromboembolism disease with clinical Galium Verum L.preparation of obviously function.
