15-PGDH, Cox-2and Beta-Catenin Relationship With The Occurrence And Development Of Colorectal Cancer

Objective: By observing the beta-catenin, cox-2and15-PGDH expressionin normal colon tissue, in adenoma and colorectal cancer tissue, exploredits role in the arcinogenesis and development of colorectal cancer.Methods: Through surgery or endoscopic examination methods, to collectspecimens of30cases of colon cancer and adenoma specimens, routinehistologic examination,10cases of normal control intestinal mucosa tissues.RT-PCR tested15-PGDH, cox-2and beta-catenin mRNA expression level innormal colon tissue, various types of colonic tumor polyps and colorectalcancer tissue. Western blot tested15-PGDH, cox-2and beta-catenin proteinexpression in normal colon tissue, various of colonic tumor polyps andcolorectal cancer tissue. By Immunohistochemical method to observe the15-PGDH, cox-2and beta–catenin in normal colon tissue and adenoma andcolorectal cancer cells. The last, the use of SPSS and IPP software analyzedexperimental results.Results: In normal colon tissue, adenomas and colorectal cancer, COX-2andbeta-catenin mRNA and protein expression was significantly higher(P <0.05),their expression in adenomas and colorectal cancer was greater than the amountof normal tissue, and the difference was statistically significant(P<0.05),there was statistically significant difference between in adenomas andcolorectal cancer(P<0.05).But15-PGDH mRNA and protein expression in thetissues was significantly reduced (P <0.05). Its expression in adenomas andcolorectal cancer was less than the amount of normal tissue, and the differencewas statistically significant(P<0.05), there was statistically significant difference between in adenomas and colorectal cancer(P<0.05).Immunohistochemical method had the same results.Conclusion: Cox-2and beta-catenin expression increase promoted thedevelopment of colorectal cancer, and15-PGDH reduce or missing promotedthe development of colorectal cancer.
