Objective: According to observe the intervening effect of1,25-(OH)2D3in type2diabetes (T2DM) rats that was modeled successfully,to explore the protective effectsand mechanisms of1,25-(OH)2D3in type2diabetic rats liver. Methods:Selectthirty-four8-week-old male SD rats that were randomly divided into control group(10),the1,25-(OH)2D3intervening group (the VD intervening group)(12), diabetic group(12).Rats in control group were given normal diet, rats in VD intervening group and diabeticgroup were fed with high fat and sugar forage. After six weeks, enterocoelia of rats in theVD intervening group and diabetic group were injected low doses of streptozotocin (STZ)to induce T2DM rat modes, select10T2DM rat modes in VD intervening group anddiabetic group to continue to feed high fat and sugar forage, rats in VD intervening groupwas given0.03ug/kg.d arachis oil which was made by1,25-(OH)2D3soluble in0.05mlarachis oil to gavage, rats in diabetic group were given the same amountpeanut of arachisoil that was not solubled in1,25-(OH)2D3to gavage, rats in control group were givennormal diet. After6weeks,measured the levels of FPG、TG、TC、ALT and AST in serumof rats in each group; eviscerate every rat,liver to weighe and calculate the hepatic index,examine the pathological morphology of liver tissue of each rat by HE staining andexamine the expression levels of TNF-alpha (tumor necrosis factor-a) and TGF-β1(transforming growth factor-β1) in each rat, liver tissue by immunohistochemistry(IHC).Results:The hepatic index、FPG、TG、TC and serum ALT and AST of rats inDiabetic group comgpared with the ones in control group were statisticallyincreasing,which were statistically significant (P<0.01), TNF-alpha and TGF-β1expression level of each rat,liver tissue in Diabetic group compared with the ones incontrol group were statistically increasing,which were statistically significant (P<0.01),theTG,ALT and AST in blood serum of rats in VD intervening group compared with the ones in the diabetic group were obviously decreasing, which were statistically significant(P<0.01),histopathological changes of rats, liver in VD intervening group compared withthe ones in diabetic group was eased. Conclusion:1,25-(OH)2D3may be formed byinhibiting fat, reducing TNF-alpha and TGF-β1protein expression to play a protectiveeffect on type2diabetic rat liver.
