ObjectiveTo investigate the expression of5-lipoxygenase(5-LOX) and c-myc protein in duodenaladenocarcinoma and paracancer tissues, we hope to know the relationship between theirexpression and the clinical significance of their expression in duodenaladenocarcinoma,at the same time, combination of follow-up data,we try to estimate therelationship between5-LOX、c-myc and the prognosis of duodenal adenocarcinomapatients with the survival analysis of duodenal adenocarcinoma patients.MethodsTo collect60cases of duodenal adenocarcinoma and30cases of paracancer tissuesfrom the Pathology of First Affiliated Hospital of Anhui Medical University,Theexpression of5-LOX and c-myc protein in60duodenal adenocarcinoma and30paracancer tissues was examined by immunohistochemical SP technique.With thefollow-up of patients,wo analyze the relationship between5-LOX、c-myc and thedevelopment、prognosis of duodenal adenocarcinoma.All data were processed by SPSS13.0.Results(1)Expression of5-LOX and the relationship between5-LOX and prognosis Thepositive expression rate of5-LOX in duodenal adenocarcinoma was significantly higher than in normal duodenal(P<0.01), And the positive expression rate of5-LOX induodenal adenocarcinoma was closely correlated with the depth of invasion,celldifferentiation and lymph node metastasis(P<0.05) but had nothing to do with age, sex,tumor size,initial occurring position.5-LOX positive expression in invaded tunicaexterna and out of tunica externa was higher than that invaded mucous membrane andmuscle(P<0.001).5-LOX positive expression in poorly differentiated cancer type washigher than that in moderately and well differentiated type(P=0.039); The expressionrate in positive lymph nodes was higher than that of negative lymph nodes(P=0.021). Inthe5-LOX positive expression group,1year、3year、5year survival rate wererespectively76.1%,26.1%,2.2%; while in negative group,1year、3year、5yearsurvival rate were respectively85.7%、42.9%、14.3%, with the Log-rank test thedifference was significant(χ2=6.626,P=0.01);(2)Expression of c-myc protein and therelationship between c-myc and prognosis The positive expression rate of c-myc induodenal adenocarcinoma was significantly higher than in normal duodenal(P<0.01),The expression of c-myc protein was correlated with the depth of invasion and lymphnode metastasis(P<0.05) but had nothing to do with other groups, c-myc positiveexpression in invaded tunica externa and out of tunica externa was higher than thatinvaded mucous membrane and muscle(P=0.015);The expression rate in positive lymphnodes was higher than that of negative lymph nodes(P=0.02).In the c-myc positiveexpression group,1year、3year、5year survival rate were respectively76.5%,27.5%,3.9%; while in negative group,1year、3year、5year survival rate were respectively88.9%、44.4%、11.1%, with the Log-rank test the difference was not significant(χ2=1.025,P>0.05);(3) Expression correlation of5-LOX and c-myc In60cases ofduodenal adenocarcinoma there is43cases both positive expression of5-LOX andc-myc,and6cases both negative expression,There is obviously correlation between theexpression of5-LOX and c-myc in duodenal adenocarcinoma(P=0.001,r=0.43). Conclusion(1)The abnormal expression of5-LOX are closely correlated with the occurrence anddevelopment of duodenal adenocarcinoma.5-LOX can promote the ability of invasionand metastasis of duodenal adenocarcinoma,which can be an non-specific index foraided diagnosis and determine the degree of malignancy of duodenal adenocarcinoma.(2) The expression of5-LOX are closely correlated with the prognosis,which can be animportant index for judge prognosis.(3)c-myc involved in the occurrence and development of duodenaladenocarcinoma,which can be an important index for invasion、metastasis and prognosisof duodenal adenocarcinoma.(4)5-LOX and c-myc are closely correlated with the occurrence and development ofduodenal adenocarcinoma,which can provide a new way for clinical treatment ofduodenal adenocarcinoma that have a good prospect.(5)5-LOX may play roles through interacting c-myc in duodenal adenocarcinoma.
