Objective: we intended to detect the expression of MMP-3 in primary duodenal adenocarcinoma,to analyze the relationship between the expression of matrix metalloproteinase 3(MMP-3) in primary duodenal adenocarcinoma(PDA) and various clinicopathologic characteristics, and to explore an important role and clinic significance which MMP-3 play in the development and progression of primary duodenal adenocarcinoma.Methods: The experiments were choose 56 cases of paraffin embedded specimens of PDA by pancreaticoduodenectomy in the First Affiliated Hospital of Nanchang University between 2010 January to 2013 December(be selected standard: 1.all patients were confirmed by pathological diagnosis;2.all patients were pancreaticoduodenectomy; 3.all patients were not preoperative radiotherapy and chemotherapy;4.clinical data of all patients’ medical record remained intact). The control group were 38 cases of normal paraneoplastic tissues of PDA by pancreaticoduodenectomy. SP immunohistochemical method was applied to detect the expression of MMP-3 in the normal tissues adjacent to cancer and the cancer tissues of PDA. Combining with clinicopathologic parameter of primary duodenal adenocarcinoma, and then we statistically analyzed the data with SPSS 17.0 statistical software and X2 inspection, P<0.05 difference has statistics significance.Results: 1.The positive expression rate of MMP-3 in cancer tissue of primary duodenal adenocarcinoma was 69.6%, which was significantly higher than that in normal paracancer tissues samples( P<0. 05).2.The expression of MMP-3 in the cancer tissue of primary duodenal adenocarcinoma were significant associated with pathological differentiation, lymph node metastasis,infiltrate depth of Cancer cells,clinical stages and Whether or not nerves were affected by cancer cells(P<0.05),but had nothing to do with the patients’ sex, age, tumor size and initial occurring position of tumor(P>0.05). 3.The degree of pathological differentiation of primary duodenal adenocarcinoma with lymph node metastasis, invasion of nerve and the depth of tumor invasion and clinical staging is closely related to(P<0.05),While the patients gender, age, tumor size, tumor location had nothing to do with tumor lymph node metastasis, invasion of nerve and the depth of tumor invasion and clinical staging(P>0.05).Conclusions: 1.The high expression of MMP-3 in primary duodenal adenocarcinoma may be closely related to tumor occurrence and development. 2.The expression level of MMP-3 may be associated with the biological behavior and prognosis of primary duodenal adenocarcinoma. 3.MMP-3 may contribute to the early diagnosis, targeted therapy and prognostic judgement of primary duodenal adenocarcinoma. 4.MMP-3 may promote the invasion and metastasis of primary duodenal adenocarcinoma, an important indicator of whether it will be able to judge the invasion force of primary duodenal adenocarcinoma as the prospective experimental study to be a large sample, multi center to further demonstrate.
