Backgrand and Objective PAKs(p21-activated kinases) belonging to the Rho-familyGTPase is Serine/Threonine kinase, containing characteristic Cdc42and Rac1interactive binding domain,which interacted with Cdc42/Rac and could be activatedby P21-GTP kinase. PAKs comprehend six isoforms PAK1-6.They overexpressed,hyperactivated or amplificated in several human tumors and they play an important rolein Cell differentiation, proliferation, viability, apoptosis, motion, gene regulation,cytoskeleton rearrangement and the generation, invasion, metastasis of cancer. PAK6was found as the protein kinase interacted with androgen receptor in2001,and itsuppressed AR-mediated transcription especially. We research the expression of PAK6in adenocarcinoma endometrium, and investigate the association of PAK6with AR, ER,PR, aiming to provide new diagnosis marker or new therapeutic notion foradenocarcinoma endometrium.Methods1. Detection the expression of PAK6, AR, ER, PR in normal endometrium,different pathology classifications and stagings adenocarcinoma endometrium byimmunohistochemistry.2. Enzyme linked immunosorbent assay was carried out todetect the concentration of PAK6in fasting blood serum of normal people andadenocarcinoma endometrium patents.Results The positive expression rate of PAK6in adenocarcinoma endometrium tissueis46.88%, while that of normal tissue is22.5%. The differences between the two groupsis statistical significant (p=0.042).The positive expression rate of PAK6in histologicalgrade G1,G2,G3is16.67%,36.36%,68%,respectively. There is statistical significance of the differences (p=0,017). And the rate in FIGO staging Ⅰ, Ⅱ, Ⅲis36.36%,50%,78.57%, analyzing the differences, p=0.002, and the result is statistical significant. Theaverage concentration of PAK6in fasting blood serum of normal people andadenocarcinoma endometrium patents is2.560±2.452ng/ml,6.197±4.233ng/ml,respectively. To analyze the differences by t test, there is statistical significance. Theexpression of PAK6is negative correlation with ER in adenocarcinoma endometriumtissue (r=-0.38, p=0.002), while there is no definite correlation between PAK6and AR,PR(r=0.17, p=0.168; r=-0.14, p=0.256).There is no relationship between theexpression of PAK6and lymph nodes metastasis, the depth of muscular infiltration,patent’s age, menopause (p>0.05).Conclusion (1) PAK6overexpressed in adenocarcinoma endometrium tissue, suggestingPAK6might play a part in the generation of the cancer.(2) The expression of PAK6increased with increasing histological gradeand FIGO stage, supposition PAK6may play a role in the development ofadenocarcinoma endometrium, and indicating PAK6may be a tissue marker forassessing pathogenetic condition.(3) PAK6high expressed in blood serum of adenocarcinoma endometriumpatents,suggesting it might be a serology index for the disease judgment and followingup after treatment.
