Background and objectiv Endometrial carcinoma is one of the three common female reproductive tract malignancy, accounted for 20% -30%. Now the pathogenesis of endometrial cancer is not very clear.In the world, the incidence of endometrial cancer is on the rise. Tumor suppressor gene inactivation and oncogene activation is an important mechanism of tumor development, and also one research hotspot of endometrial carcinoma. In this study, by detecting the protein expression of PTEN, p16 in endometrial tissue, to explore the relationship of genesis and development and the significance in endometrial carcinoma, and the possibility of early diagnosis and judging the prognosis in endometrial carcinoma.Method This experiment uses immunohistochemical streptavidin-biotin-perxida--es conjugated (streptavidin-perosidase SP) method, to detecte the expression level of PTEN, p16 protein in the 20 example normal endometrium, the 40example organization of endometrial hyperplasia and the 31 example endometrium adenocarcinoma organizes. Combining clinical and pathological information, according to histological classificati--on, we divide the cancer issues into high, medium and low groups, according to the 2000 International Federation of Gynecology and Obstetrics (FIGO) criteria, we divide the cancer issues into surgical staging group (â… -â…£period a total of four), Meanwhile, in order to judge the prognosis in endometrial carcinoma, we list the lymph node shift whether or not and the myo-level infiltration especially (in 1/2 for the sector), we use the microscope for cell counting, and the SPSS13.0 statistics software for the data.Result1. The positive expression rate of PTEN protein was decreasing trend in the normal endometrium group, endometrial hyperplasia group(simple hyperplasia, complex hyperplasia, atypical hyperplasia), and endometrial adenocarcinoma group(P<0.05);the expression of PTEN protein in normal endometrium, complex hyperplasia and atypical hyperplasia was significantly different(P<0.05); the expression of PTEN protein in endometrial adenocarcinoma and complex hyperplasia tissue was significantly different (P<0.05).2. The expression of p16 protein in endometrial adenocarcinoma and normal endometrium and endometrial hyperplasia group was significantly different(P<0. 05);the expression of p16 protein in normal endometrium and endometrial hyperplasia was no significant difference(P>0.05) ;there was no significant difference among the endometrial hyperplasia(P>0.05).3. The expression of PTEN p16 protein have a ralation in endometrial carcinoma and surgical staging histological grade, degree of differentiation(P<0.05).4. The expression of PTEN, p16 protein in endometrial carcinoma tissues was positively correlated( r = 1, P < 0.05).Conclusion1. The expression of PTEN in normal endometrium and endometrial atypical hyperplasia and endometrial adenocarcinoma group was decreasing trend, this prompted that PTEN was associated with the occurrence of endometrial adenocarcinoma and the development process, it may provides a biological target of an early diagnosis in atypical hyperplasia and endometrial adenocarcinoma.2. The expression of p16 in endometrial adenocarcinoma group was significantly decreasing, it may be associated with endometrium lesion extent.3. The expression of PTEN and p16 have a ralation in endometrial cancer of the pathological stage of the surgery, degree of differentiation, this prompts the both may be associated with tumor's biological behavior of invasion, metastasis and so on.4. PTEN and p16 play a role of synergism at the endometrium adenocarcinoma formation and the biology behavior change process, possibly promoting tumour cell's multiplication and the differentiation, and may become important targets of judgeing endometrium adenocarcinoma prognosis.
