ObjectiveTo study the expression of monocyte chemotactic factor-1(MCP-1) and fibronectin (FN) in human secondary acquired middle ear cholesteatoma epithelium, and to investigate the correlation between their expression and the ability of erosion of cholesteatoma.MethodsMaxVisionTM immunohistochemical method was used to detect the expression of MCP-1and FN in the secondary acquired middle ear cholesteatoma tissues from30patients and in the retroauricular skin from20patients and in the retroauricular skin from16normal subjects. Then positive values of cells where measured by manual counting method and image analysis technology. All the dates were analysised by SPSS17.0, P<0.05was refered to statistically significant difference.ResultsMCP-1appeared to local in all epithelial layers of middle ear cholesteatoma, particularly in the spinous layers. The positive expression rate of MCP-1was70%, the gray value was147.2±20.1, Which were siginificantly higher than in the retroauricular skin from patients (35%.200.8±18.4) and from normal subjects (37.5%,193.3±15.5)(P<0.05). The expression of FN in all epithelial layers of middle ear cholesteatoma were abundantly stained, especially in the basal and spinous layers and the matrix of cholesteatoma. The positive expression rate of FN was76.7%, the gray value was147.2±20.1, Which were siginificantly higher than in the retroauricular skin from patients (30%.195.0±12.9) and from normal subjects (31.3%,191.6±13.5)(P<0.05). It showed statistically significant correlation between the expression of MCP-1or FN and the ability of erosion of middle ear cholesteatoma(rmcp-1=-0.682, rfn=-0.531, P<0.01). There was not linear correlation between the expression of MCP-1and FN. Moreover, The expression of MCP-1in the fourth level of bone destruction was weaker than in the previous three levels (P<0.05). The expression of FN was not significant difference in the four levels of bone destruction (P>0.05)ConclusionMCP-1and FN overexpressed in middle ear cholesteatoma.There was correlation between the expression of MCP-1or FN and the ability of erosion of middle ear cholesteatoma, And,MCP-1and FN sustained expression in four levels of bone destruction, Which shows that MCP-1and FN might play an important role in invasive behavior of cholesteatoma.
