| Objective:Cholesteatoma Otitis Media destroys tympanic structure.It can cause the serious complications of intra and extra cranial.The aim is to investigate the mechanism of bony destruction in Cholesteatoma Otitis Media by the OPN expression on Cholesteatoma epithelium.Methods:According to the standards,we selected 25 cases of cholesteatoma otitis media patients as experimental group,collected 25 surgical resection specimens,and at the same time to select 15 cases of bone with external auditory canal skin in these patients as a control group,collected 15 normal skin samples of the external auditory canal.All specimens required to formalin-fixed,paraffin-embedded.Each of the experimental specimens were done with hematoxylin and eosin staining to determine the pathological diagnosis.Two-step immunohistochemical method was used to detect the expression of OPN on epithelium of 25 cases with Cholesteatoma Media and on the skin tissue of bony portion for 10 cases with normal external auditory canal.SPSS13.0 system was used to perform the statistical analysis.Results:17 of 25 cases of Cholesteatoma showed strong positive expression on basal cell layer,prickel cell layer and stratum granulosum of cholesteatoma epithelium,while 2 cases showed moderate positive,they were positively pigmented in the cytoplasm.6 of 25 cases showed negative expression,10 cases with normal skin tissue on bony portion of external acoustic meatus were all negative expression.Conclusions:The expression of OPN in cholesteatoma otitis media is significantly higher than the skin tissue of the bony portion with the external auditory canal,suggesting that OPN may be a correlation of bone destruction in Cholesteatoma. |
