Objective To investigate the gastrodin given the impact on rat’s postoperative cognitive function with propofol anesthesia and their relationship of hippocampal nerve growth factor (NGF)、brain derived neurotrophic factor (BDNF) variation in content, providing a reference for clinical.Method Sixty-four healthy male SD rats (body weight was200±20g,2months of ages) were randomly divided into4groups:normal group (N), propofol group (P), abdominal operation group (S), gastrodin group (G),16rats in each group. Propofol group intraperitoneal injection of propofol100mg/kg anesthesia (10ml/kg) for2hours. Operation group and gastrodin group100mg/kg propofol anesthesia (lOml/kg) in the process for exploratory laparotomy,2hours of continuous time; gastrodin group after continuous intraperitoneal injection of gastrodin100mg/kg (1ml/kg)7days. Normal group received an equal volume of saline. After3,5and7days respectively, Morris water maze experiment rat spatial learning and memory ability,2hours after the end of the behavioral test,killing the rats to get out the hippocampus, hippocampus part of extraction of total RNA detected by RT-PCR NGF and BDNF mRNA expression. Parts of hippocampus were fixed by paraformaldehyde after paraffin section, immunohistochemical staining of hippocampal NGF and BDNF expression.Results1.Morris water maze test resultes:①Place navigation test:Preoperative4group is generally not statistically significant (P>0.05); there was statistical significance after3days compared with the P group, S group and G group and N group differences in latency (P<0.05), P group, S group and G group than in N group was significantly prolonged the latency; after3days,5days,7days comparison of S, G group, P group and N latency difference was statistically significant (P<0.05), S, N, G group was longer than that in group P was significantly prolonged;7days after operation in group G than in group S was significantly shorter latencies (P<0.05);3days after operation, N group, P group, S group and G group latency the preoperative and the difference was statistically significant (P<0.05);5days after operation, group N latent period and preoperative difference was statistically significant (P<0.05), there was statistical significance in P, S, G group after3days incubation with difference (P<0.05); after7days before operation, four groups and3days after surgery were statistically significant (P<0.05), there is statistical significance after7days in P group, S group and G group after5days of incubation period and the difference (P<0.05)..②Spatial probe test:a. Before operation,4groups of rats of the percentage of overall residence time had no statistical significance (P>0.05); after3days of the original platform residence time percentage of P group, S group, G group and N group, the difference was statistically significant (P<0.05), the original platform residence time percent of N group decreased significantly, while the group S, group G of the residence time of percentage reduction of more (P<0.05), compared with the P group; there was significant difference after5to7days, group S, group G platform and N, the percentage time spent in P group (P<0.05), the residence time of N, percent of group P decreased there are significant differences;7days after operation in group S platform the percentage time spent with G group (P<0.05), G group of the residence time of S group was significantly increased percentage.b. Before operation,4groups of passing through the platform number no statistical difference (P>0.05); there were significant differences in postoperative3days P, S, G group and N group (P<0.05), P, S, G group of passing through the platform number was less than that of N, S, G group of passing through the platform number less than P group (P<0.05);5days after operation, S, G group through the comparison of the original number of platform and N, P groups were statistically different (P<0.05), S, G group of passing through the platform number was less than N, P group;7days after operation, S group through the comparison of the original number of platform and N, P and G groups there was a statistically significant difference (P<0.05), S group through the times in platform is N, P and G decreased.2. The results of RT-PCR detection:3days after operation, P, S and G in group NGF, BDNF mRNA than in group N appeared to be down-regulated significantly (P<0.05), and the expression of S and down-regulated in the G group was more significant, there are significant differences compared with the P group (P<0.05); and after3days,7days P, S and G groups in NGF, BDNF mRNA expression was significantly increased after operation (P<0.05); compared with N group, P group after7days of hippocampal NGF, BDNF mRNA expression had no statistical significance (P>0.05); compared with S group, P group and G group for7days in NGF, BDNF mRNA expression were higher than that in group S after operation (P<0.05), and P group increased more significantly, there is significant compared with group G (P<0.05).3. Results immunohistochemistry:Compared with group N, the average gray of positive cells in hippocampus CA1area of NGF group and BDNF P values were not statistically significant (P>0.05); compared with P group, the average gray of NGF positive cells and BDNF S and G in hippocampus CA1area of low value (P<0.05), while S was lower, with a significant difference in comparison group G (P<0.05).Conclusion1.Propofol anesthesia can lead to the decrease of the ability of learning and memory in the short term space of normal rats, with the extension of time this effect decreases gradually. Noxious stimulation of operation can further aggravate the propofol anesthesia in the cognitive function of the rats with damage.2. Rats after long-term use of gastrodin treatment, can significantly improve the learning and memory ability of propofol anesthesia in rats after space.3. The hippocampus endogenous NGF, BDNF increased in the rat spatial learning memory function recovery process.4. Gastrodin can increase the expression of endogenous NGF, BDNF in hippocampus, and enhanced spatial learning and memory ability in rats, promote the postoperative cognitive function improvement.
