Objective:To explore the effects of propofol on the cognitive function in rats with laparotomy, and Panax notoginseng saponins intervention whether has the role to improve cognition or not.Method:64Male SD rats weighing180-220g were randomly divided into four groups:normal group (n=16); propofol group (n=16); surgical group (n=16); PNS group (n=16). The rats receive propofol100mg/kg (10ml/kg) by intraperitoneal injection firstly, and were boosted one-half of the initial dose when movement emerged for2hours. Assessing the cognition of rats with Morris water maze at3d and7d after the operation. To do the RT-PCR at3d and7d after surgery, and to do the immunohistochemistry at7d after surgery. We can detect BDNF\SYP expression in the Hippocampus of brain by RT-PCR and immunohistochemistry.Results:1. Morris water maze results:①Place Navigation:compared to the normal group, propofol group, surgical group and PNS group’s latency were increased, the differences were statistically significant (P<0.01); compared to the surgical group, propofol group and PNS group’s latency were reduced, and the differences were statistically significant (P<0.05); but the difference between propofol group and PNS group was not statistically significant (P>0.05). Propofol and PNS groups in3d postoperative, latency were both increased than1d preoperative and7d postoperative, the difference was statistically significant (P<0.05), but the difference between1d pre-and7d postoperative was not statistically significant (P>0.05). Compared to Id preoperative, surgical group in3d and7d postoperative, the difference was statistically significant (P<0.05).②Probe Trials:Compared to normal group, the other three groups’ frequencies of crossing platform were reduced in3d and7d postoperative, the differences were statistically significant (P<0.05);7d postoperative, compared with the surgical group, propofol and PNS groups’frequencies were increased, and the difference was statistically significant (P<0.05).2. RT-PCR results:①Bdnf:compared with the normal group, mRNA expression of propofol group was reduced in3d and7d postoperative, but the difference was not statistically significant (P>0.05), but the surgical and PNS groups’was decreased, and the difference was statistically significant (P<0.05);②Syp:compared with the normal group, mRNA expression of propofol group was decreased in3d postoperative, the difference was statistically significant (P<0.05), but the expression’s difference was no significant in7d postoperative(P>0.05); surgical and PNS groups’expression after were decreased in3d,7d postoperative, and the difference was statistically significant (P<0.05); PNS group’s expression was increased in7d postoperative to compared with3d postoperative, and the difference was statistically significant (P<0.05).3. IHC results:①BDNF:propofol group’s hippocampus nerve cell cytoplasm was stained in brown yellow deeply, and the staining intensity match with the normal group in7d postoperative, but the surgical and PNS groups’staining were not obvious.②SYP:compared with the normal group, propofol group’s synaptophysin of neuropil was stained into brown yellow granular in7d postoperative, and was lighter than the normal group’s; surgical and PNS groups’ were stained into pale yellow granular; and PNS group’s was darker than surgical’s.Conclusion:Propofol can lead to cognitive dysfunction in rats in the short term, this may be related to the decreased SYP expression in hippocampus; the decreased BNDF expression in hippocampus may caused by surgical stimulation, and this could be one of the factors that cause postoperative cognitive dysfunction; PNS can promote the SYP expression in hippocampus, which may improve postoperative cognitive function in rats, but the specific pathway remains to be studied.
