Objective:To observe propofol on cognitive functions such as spatial learn ing, memory and the influence of the hippocampus on hyperlipidemia seni le rat. Methods: 76 aged male SD rats were selected and randomly divid ed into basal diet with propofol injection(group B1), basal diet with nor mal saline injection(group N1), high-fat diet with propofol injection(group B2), high-fat diet with normal saline injection(group B2),each group of 19. After fed respectively 8 weeks, group B1 and B2 received intraperiton eal injecting of propofol 100mg/kg/d for 5 days. While in group N1 and N2, rats received the same intraperitoneal injecting of equal volume norm al saline. One day after the last injection, escape latency time and space exploration were detected with Morris water maze in the next 6 days. 1 hour after the last water maze test, the aged rats serum and hippocampus were sampled to detect the expression of beta-amyloid protein and RAG E by means of immunohistochemical and ELISA. Results: In place navig ation test B1’s escape incubation period(98.20±25.40) was obviously longer than N1(47.50±11.08)(P<0.01). Compared with B2(99.79±20.38),escape incubation period was shortened in N2(50.70±20.55)(P<0.05). There were no significantly difference between N2 and N1(P>0.05), whi le B2’s escape incubation period was longer than B1(P<0.05). In spatial probe test, platform number(2.86 ±1.46) in group B1 was less than N1 group(7.5±1.70)(P<0.05). In group N2, the platform number(6.60 ±3.91) was more than group B2(1.16 ±1.16)(P<0.05), there was no statistic ally significant difference between group N1 and N2(P>0.05).Times of c rossing platform in group B2 were less than that in B1 group(P<0.05). In Elisa, compared with control group N1[(147.83±60.88)ng/L] and N2[(152.73±87.50)ng/L], expression of RAGE protein was increased in group B1[(629.89±110.33)ng/L] and B2[(229.89±53.20)ng/L](P<0.05). There was no significantly difference between N1 and N2 group(P>0.05), while expres sion of RAGE protein in B1 group was lower than B2(P<0.05). In i mmunohistochemical test, positive expressing cells counting in B1(18.49±1.53) and B2 respectively(25.67±3.08) is higher than(9.33 ±2.31)group N1 a nd(12.14±2.52) group N2(P<0.05), there was no statistically significant difference between group N1 and N2(P>0.05). Conclusions:Anesthetic do ses of propofol can injury spatial learning and memory ability of the seni le rat, resulting in the decreasing of cognitive function in aged rats. Hype rlipidemia might act synergistically withpropofol.
