Objective: Detection the infection of Hp in chronic atrophic gastritis (CAG), gastrohelcosis (GH),gastric intraepithelial neoplasia (GIN), gastric cancer (GC) and adjacent tissues of cancer (AC),and the expression levels of IL-17A and IL-17F in gastric cancer and adjacent tissues, respectivelydiscussion their correlation and the clinical pathological significance on the gastric cancer’mechanism between Hp, IL-17A and IL-17F.Methods:(1) Selected181cases of gastric paraffin-embedded; collected104cases of AC,79cases of them are NM and the rest of25cases are GIN; in addition,2054cases of glass slice arestained by the methylene blue, of which1638cases of CAG,238cases of GIN,178cases of GH;there are10cases of fresh gastric cancer tissues; paraffin-embedded tissues were fixed by theformalin.(2) Methylene blue staining and immunohistochemistry were used to detect the groups’Hp infection and the groups’ expression of IL-17A and IL-17F; detected their genes and proteins’expression levels of IL-17A and IL-17F in gastric cancer fresh tissue by RT-PCR and Western Blotmethods.(3) All datas were analyzed by SPSS17.0statistical package.Results:(1) Respectively, the Hp infection rates in the CAG, GIN, GH and GC groups were39.6%,55.0%,55.1%,66.9and73.1%. Comparison with the rest of the groups each other, thedifference was statistically significant (P<0.05). Compared HP-positive infection rates betweenAC (NM GIN) and GC, the difference was not statistically significant (χ2=0.143, P>0.05).(2) Inthe GC group IL-17A and IL-17F’positive and in the middle-strong positive expression rates wassignificantly higher than that in the control group (NM, GIN), the differences were statisticallysignificant (P<0.05); at the IL-17A and IL-17F’ Genes and protein’ expression levels, GC wassignificantly enhanced than AC, and the late (III+IV) GC was also significantly enhanced thanearlier (I+II) GC; in GC tissues, IL-17A and IL-17F’middle-strong positive rates were positivelyrelated with its depth of invasion, lymph node metastasis, distant metastasis, TNM stage (P<0.05),regardless of their ages, and genders (P>0.05).(3) It showed a positive correlation that thesurvival rate of the GC patients was related on the age, clinical stage, depth of invasion, lymphnode metastasis, distant metastasis, Hp infection, the expression of IL-17A and IL-17F (P<0.05),regardless of the gender and degree of differentiation (P>0.05). In addition, their ages, clinicalstages, depth of invasion, lymph node metastasis, distant metastasis can increase the risk of deathin patients.Conclusions:(1) In CAG, precancerous lesions (GH, GIN) and GC groups, Hp infection ratesincreased in turn, it suggested that Hp may be involved in the process from normal gastric mucosainto gastric.(2) IL-17A and IL-17F’ highly expressions in GC tissues and their positive correlationwith Hp infection, it suggested that Hp may longstand in chronic inflammation and may cause thecontinuous release of inflammatory cytokines, leading to the occurrence and, development ofgastric cancer.(3) In GC tissues, the high expression of IL-17A and IL-17F and Hp-positive infections, can significantly reduce the survival rate of patients and the median survival time,suggesting that IL-17A and IL-17F can be used as an indicator of the prognosis of GC, and Hpalso need to line eradication therapy.
