Gastric cancer is a malignant mass lesion topped various malignanttumors in China, which occurs in elderly patients, and after the age of50, theincidence is significantly higher. The incidence of male and female is2:1, butthe disease is insidious onset high degree of clinical malignancy, the prognosisis poor, and is an important health threat bote in China and the world. Thecurrent study is one of the molecular biology of tumor, which has awarenessthat it is an important method of treatment to gastric cancer, in which tumorsthe study of molecular biology research on nuclear transcription factor kappaB (Nuclear transcription factor κB, NF-κB) is a research focus of many one,and has come to realize the NF-κB is correlated with a variety of solidtumors, and can modulate inflammatory cytokines, vascular apoptosis andtumor cells, lymphangiogenesis in tumor occurrence and development of avariety of the role of the current research on NF-κB in a variety of cancersmore, but modern biological research has shows that its expression issignificant in different clinical stages of the gastric mucosa and its relationshipwith Ki-67, the relationship between TOPO II study of rare, It has beenconfirmed that Ki-67, TOPO II with a variety of benign and malignant tumors(including gastric cancer and other tumors) is closely related to furtherexplore the relationship among the three and biological behavior of gastriccancer and between the has some clinical significance.NF-κB is not only involved in the development of a variety of tumors byNF-κB pathway, while there is a clear relationship between the biologicalbehavior of gastric cancer chemotherapy affect treatment outcomes, drug, etc.for gastric cancer research, prevention and treatment of clinical biologicalbehavior and prognosis assessment and provide a theoretical basis. Objective:By studying NF-κB, Ki-67, TOPO II in gastric tissue in different clinicalstages, understanding the relationship between their expression and biologicalbehavior of gastric cancer in gastric cancer between order for the biologicalbehavior of gastric cancer, provide the basis for assessing prognosis.Method:1. Cases of gastric cancer study All gastric adenocarcinoma specimenscome from Chengde Central Hospital in2011and2013surgical oncologypatients, male72cases, female30cases, aged frome42to78years, medianAge59years, aged≥55years old in69cases,<55years,33cases were notreceived chemotherapy before surgery; pathologically confirmedadenocarcinoma, moderately differentiated adenocarcinoma,33cases ofpoorly differentiated adenocarcinoma69cases; not invading the serosa ofgastric carcinoma were36cases of gastric cancer were66cases of serosalinvasion, lymph node metastasis of gastric cancer specimens not42cases,there are60cases of gastric cancer specimens were transferred prompt, selectanother paracancerous hyperplasia organizational groups (from5cm-10cmtumor foci between adjacent hyperplasia as cancer),45cases of normal gastricmucosa (10cm from the tumor foci outside as normal gastric mucosa)organizers of34cases as a control study.2. Handling specimens collected specimens of Chengde Central Hospital,pathological2011-2013oncology patients after surgery paraffin blocks of102confirmed cases of gastric adenocarcinoma, hyperplastic tissue adjacentnormal gastric tissue removed organizations should try to prevent it frombending, torsion, should be flat on the subsequent adhesion and slowly placedin10%neutral formalin solution fixed the toilet paper, fixative is10times theamount of the fixed tissue, tissue fixation time12hours, embedded in paraffin,4μm serial sections, after dewaxing, de-xylene, after hydration usingimmunohistochemistry method streptomycin avidin-peroxidase(streptavidin-peroxidase, SP) method Test specimens in NF-κB, Ki-67, theexpression of TOPO II to PBS instead of primary antibody negative control, all reagents are instruments reagents, all operations are in strict accordance with the instrument and kit instructions, by two disease MD, deputy directorof science at the microscope more than confirmed its expression level ingastric cancer tissues and normal gastric mucosa, combined with pathologicaldata were statistically analyzed experimental data.Result:(1)102cases of gastric cancer tissue specimens, Expression of NF-κB inpositive tissue is72cases, the positive rate was70.59%; beside the45cases ofcancer hyperplasia positive expression group was10cases, the positive ratewas22.22%;34cases positive expression in normal tissues as two cases, thepositive rate was5.88%, hyperplastic tissue and adjacent normal gastricmucosa of NF-κB expression was significantly lower than that of gastriccarcinoma. The chi-square test, there are differences (2=57.104, p <0.05)between the three statistical results.(2)102cases of gastric cancer tissue specimens, expression of TOPO Ⅱ in55cases, the positive expression rate of53.92%, next to the45cases of cancerhyperplasia expression in24cases, the positive rate of53.33%, the positiveexpression in34cases of normal gastric mucosa in10cases, the positive rateof29.41%, by chi-square test, there are differences (2=6.281, p <0.05)between the three statistical results.(3)102cases of gastric cancer tissue specimens, Ki-67expression in88cases,the positive expression rate of86.27%, next to the45cases of cancerhyperplasia expression in31cases, the positive rate of68.89%,34cases ofnormal gastric mucosa expression in11cases, the positive rate32.35%,chi-square test, there are differences (2=36.83, p <0.05) between the threestatistical results.(4) Three indicators of the cases of positive expression in gastric carcinomawere72cases,55cases and88cases, there are differences between thestatistical results of the chi-square test prompted three (2=25.556, p <0.05),further dividing line chi-square test, there are significant differences betweenthe three pairwise. Conclusion:1NF-κB expression in different stages of the mucosa have statisticallysignificant in lymph node metastasis, depth of invasion and clinical stage andgradually increasing deterioration of the specimen, the positive expression wassignificantly increased, suggesting that NF-κB and clinical gastric cancerphases are closely related.2In normal gastric mucosa and precancerous lesions, we detected that theexpression of NF-κB, suggest that NF-κB may still be involved in gastriccancer and may paly a role in the starting of cancer; addition, mucosal lesionswith the deterioration of the progress of the process, Expression of NF-κB wasgradually increasing trend, suggesting that NF-κB might be involved in theprogression of gastric mucosa in various stages of damage.3TOPO Ⅱ and Ki-67was expressed in different tissues staging of gastricmucosa, and the positive expression rate increases with deepening andexpression of NF-κB in gastric malignancy while gradually increasing trend, tips and NF-κB expression in the presence of relevant sex.
