Objectives: To detect the promoter of CCNA1gene methylation in cervical cancer tissues of Uigur womenin Xinjiang, to understand the role of CCNA1gene in carcinogenesis and develop-ment of cervical cancer.Methods:1. collect110cervical specimens of Uigur women in Xinjiang (40cervical cancer tissue;40cervical normal tissues;10CIN1,10CIN2and10CIN3tissues).2. Extract the genomic DNA of the110specimens respectively.3. HPV16and HPV18infection of each specimen were detected by PCR method.4.The promoter of CCNA1gene methylation status of each specimen were detected by MS-PCR method.5.CCNA1gene expression status of each specimen were detected by RT-PCR method.6. the chi-squarestatistical analysis.Results:1. In cervical cancer, CCNA1gene promoter region methylation was87.5%, while in cervicalnormal tissues, CCNA1gene promoter region methylation was2.5%, the difference was statisticallysignificance (X~2=58.4, P <0.05).2, In the40cases of cervical normal tissues, HPV16infection rate was12.5%(5/40),40cases of cervical carcinoma tissues, HPV16infection rate of55%(22/40), the differencewas statistically significance (X~2=16.2, P <0.05);3. In40cases of normal tissues, HPV18infection rate of0%(0/40),40cases of cervical carcinoma tissues, HPV18infection rate was7.5%(3/40), the differencewas not statistically significance (X~2=1.39, P>0.05).4. The comparison results of methylation rates of alllevels of cervical neoplasia by Fisher’s exact test. Between CIN1and CIN2methylation rate difference wasnot statistically significant (P>0.0167); Between CIN3and CIN2methylation rate difference was notstatistically significant (P>0.0167); Between CIN1and CIN3methylation rate difference was statisticallysignificant (P <0.0167).5CCNA1gene mRNA expression level is higher in cervical normal tissues thanthose cervical carcinoma tissues by RT-PCR method.6.CCNA1methylation and HPV16infections wasunrelated statistically significant in cancer tissue,(P>0.05).Conclusion:1.The promoter of CCNA1gene methylation is related to tumorigenesis of cervical squamouscell carcinoma of Uigur women in Xinjiang.2.CCNA1gene mRNA expression level reduced in cervicalcancer is associated with the promoter of CCNA1gene methylation.3. The promoter of CCNA1genemethylation is not related with HPV16infection in cervical carcinoma. |
