Objective: To determine the distribution of p21Ser31Arg, p53Arg72Pro polymorphism and HPV16 DNA and correlate these two polymorphism to genetic susceptibility in HPV -associated cervical carcinoma in Uigur women in Xinjiang. To detect expression of P21 and P53 and explore the carcinogenesis of cervical carcinoma.Methods: PCR ,semi-nested PCR and PCR-RFLP were performed to detected the genotypes distribution of p21Ser31Arg, p53Arg72Pro and HPV16DNA in 152 cases of cervical carcinoma and 110 cases of normal cervical tissue in Uigur women in Xinjiang. Immunohistochemistry was used to detect the expression of P21 and P53 in 79 cases of cervical carcinoma and 40 cases of normal cervical tissue.Results: (1) frequencies of Arg/Arg, Ser/Ser and Arg/Ser in p21 were 13.0% (13/100), 38.0% (38/100) and 49.0%(49/100) in 100 cases of cervical carcinoma and 10.0%(10/100), 39.0%(39/100) and 51%(51/100) in control cases, respectively. Difference of genotype frequencies between the cervical carcinomas and the controls was not significant statistically (χ~2=0.444 , P=0.801).(2) frequencies of Arg/Arg, Pro/Pro and Arg/Pro in p53 were 46.1%(70/152), 13.1% (20/152) and 40.8%(62/152) in 152 cases of cervical carcinoma cases and 30.9%(34/110), 14.54%(16/110) and 54.55%(60/110) in control cases, respectively. Difference of genotype frequencies between the cervical carcinomas and the controls was significant statistically (χ~2=6.307,P=0.041).(3) In cervical carcinoma, the frequency of HPV16DNA of detection was 74.3%(113/152), which was significantly higher than those in control group 3.6%(4/110).Difference of HPV16 positive rate between the cervical carcinomas and the controls was significant statistically (χ~2=126.25,P=0.000) .(4) The positive rate of P21 protein was 96.2%(76/79) in cervical carcinoma, which was higher significantly than those in control group 35.0%(14/40).
