The Study Of The COL-Ⅱ And MMP-13Expressed In The Rabbit Knee Joint Cartilage And The Clinical Effect Observation By Yi Qi Yang Xue Granule

Objective:This subject discusses the express of MMP-13and COL-II in cartilage cells Yi Qi Yang Xue granule. Revealing its parts pharmacological effect and mechanism of action to remedy the osteoarthritis (OA). Through clinical research, discussing the clinical efficacy and safety of Yi Qi Yang Xue granule cure osteoarthritis. To provide the theory basis of knee osteoarthritis cure.Methods:1.40rabbits randomly divided into4groups, the sham group,the KOA m-odel group, the positive group(liu pu an) and the Yi Qi Yang Xue granule group.All groups set up the model By the method of improved Hulth right KOA. after drug in tervention by6weeks, using HE staining to observe the cartilage degenerationd and Immunohistochemical detection of the expression of COL-Ⅱ and MMP13in cartela ge.2. The clinical experiment of this subject selected60cases of acute osteoarthritis patients. Then divided them into treatment group and control group randomly accord-ing to random number table method,each group has30patients. After4weeks, obse rved the changes of symptoms,signs,symptom integral,the blood routine the urinerou tine, ESR,CRP andSF-36of patients.Results:1. Experimental studies have shown that:the hematoxylin-ensin staining in yiqiyangxue granule group and positive medicine group compared with model group, the knee joint cartilage surface is relatively smooth,cell line relatively clutterfree, thi-nning of the cartilage layer is lighter. The express of cartilage cells MMP-13and COL-Ⅱ in yiqiyangxue granule group and positive medicine group of is significant differences (P<0.01). But no significant difference between two groups (P>0.05) 2.Two groups of Chinese medicine symptom integral,the WOMAC index integral, SF-36and CRP were improved(P>0.05),Two groups of liver kidney function,bl-ood ro-utine urine during this experiment observed adverse effects were found (P>0.05)Conclution:1. Yi Qi Yang Xue granule can protect the rabbit knee joint cartilage, we conclude that it may adjust the expression of MMP-13, COL-Ⅱ in OA cartilage cell-s, to delay the degeneration of articular cartilage.2. Yiqi yangxue granule is safe and effective drug for treatment of knee osteoarthritis. It deserves further study.
