[Objective]Based on the TCM hour theory of “the symptoms of diseases are relieving inthe morning, remaining stability during daytime, increasing at nightfall andbecoming serious at midnight” and “lung channels are vigorous during the period of3a.m. to5a.m.”, discuss the curative effectiveness of the combination of lung cancerdecoction I and cyclophosphamide treating lung cancer of mouse Lewis in differenthours. In order to provide experimental references for the combination of TCM andwestern medicine, explore the way of experimental research on the combination ofTCM and western medicine.[Methods]Copy the mode of mouse Lewis with lung cancer, dividing the mouse Lewiswith lung cancer into four groups: bearing cancer control group, treatment group of9p.m. to11p.m., treatment group of3a.m. to5a.m., treatment group of9a.m. to11a.m. There is69minutes’ time difference between Kunming time and Beijing time,therefore, the time for giving medicine to treatment group of9p.m. to11p.m.,treatment group of3a.m. to5a.m., treatment group of a.m. to11a.m. are22:09-00:09,04:09-06:09and10:09-12:09respectively.Except the bearing cancer control group, the mouse Lewis with lung cancer ofother3groups are given Lung Cancer Decoction I once in different hours per day,intraperitoneal injection of Cyclophosphamide once in different hours every3days.Then test the water and food volume of the mouse and observe their change ofmovement. Kill the mouse to get specimen after taking medicine12days, testing thetelomerase activity and apoptosis rate and making histopathologic examination. [Results]1. After getting vaccinated6days, we can see some tumor under the right forelegoxter and subcutaneous of the mouse, which indicate the mode has been achieved.2. Tumor weight and anti tumor rate: the average tumor weight of each treatmentgroup is lower than the bearing cancer control group, the difference has statisticalsignificance (P<0.01), the average tumor weight of treatment group of9a.m. to11a.m.is lower than treatment group of9p.m. to11p.m. remarkably, the difference hasstatistical significance (P<0.01), the average tumor weight of treatment group of9a.m.to11a.m. is lower than treatment group of3a.m. to5a.m. obviously, the differencehas statistical significance (P<0.01), the tumor weight of treatment group of9p.m. to11p.m. is inferior to treatment group of3a.m. to5a.m, the difference has no statisticalsignificance(P>0.05). For the anti tumor rate, the anti tumor rate of treatment group of9a.m. to11a.m. is63.39%, treatment group of9a.m. to11a.m. is39.16%, treatmentgroup of3a.m. to5a.m is33.52%, treatment group of9a.m. to11a.m. has thestrongest impact on anti tumor.3. Testing the telomerase activity: the Ct value of treatment group of9a.m. to11a.m. is higher than bearing cancer control group, the difference has statisticalsignificance (P<0.01), the Ct value of treatment group of9p.m. to11p.m. is higherthan bearing cancer control group, the difference has statistical significance (P<0.01),the Ct value of treatment group of3a.m. to5a.m is higher than bearing cancer controlgroup, the difference has statistical significance (P<0.01). Comparing the3treatmentgroups, there is no significant difference among them, the difference has no statisticalsignificance (P>0.05).4. Testing apoptosis rate: the apoptosis rate of treatment group of9a.m. to11a.m. is higher than bearing cancer control group, the difference has statisticalsignificance (P<0.01), the apoptosis rate of treatment group of9p.m. to11p.m. ishigher than bearing cancer control group, the difference has statistical significance(P<0.01), the apoptosis rate of treatment group of3a.m. to5a.m is higher than bearingcancer control group, the difference has statistical significance (P<0.01). Theapoptosis rate of treatment group of9a.m. to11a.m. higher than treatment group of9p.m. to11p.m., the difference has statistical significance (P<0.05), the apoptosis rate of treatment group of9a.m. to11a.m. higher than treatment group of3a.m. to5a.m,the difference has statistical significance (P<0.05), there is no significant differencebetween treatment group of9p.m. to11p.m. and treatment group of3a.m. to5a.m,no statistical significance (P>0.05).5. Observing the histology: the cells of each group are different under themicroscope, the cell nucleuses are big and anachromasis, has strong atypia, which canbe confirmed as tumor cells. Compared the bearing cancer control group to othertreatment groups, the cancer cell density decreased, thanatosis area increased, cellsinfiltrated by interstitial substance increased, while there is no significant differenceamong treatment groups.[Conclusions]1. Comparing the index of bearing cancer control group with the treatmentgroups, testified the curative effectiveness of the combination of lung cancerdecoction I and cyclophosphamide treating mouse Lewis with lung cancer.2. The index of tumor weight, anti tumor and apoptosis rate of treatment group of9a.m. to11a.m. is superior to treatment group of9p.m. to11p.m. and treatmentgroup of3a.m. to5a.m, proposed that the TCM hour theory of “the symptoms ofdiseases are relieving in the morning, remaining stability during daytime, increasing atnightfall and becoming serious at midnight” has certain guiding role in the treatmentof combining TCM and western medicine.3. Each index of treatment group of3a.m. to5a.m is better than the treatmentgroup of9a.m. to11a.m. and treatment group of9p.m. to11p.m., indicated that theTCM hour theory of “lung channels are vigorous during the period of3a.m. to5a.m.”has no obvious influence on the mouse modes with lung cancer vaccinated bysubcutaneous.4. Adopt the TCM hour theory to guide the combination of TCM and westernmedicine, explore the way of combining TCM and western medicine through theexperiment.
