Objective To investigate the Hui medicine AiKangFang mice Lewis lung tumor cellgrowth inhibition and of p53, Bcl-2gene expression.Methods There were55Lewis lung tumor-bearing C57BL/6J mice were divided intotumor-bearing model group, cyclophosphamide (CTX) group,Hui medicine AiKangFanglow-dose, Hui medicine AiKangFang low-dose+CTX group, Hui medicine AiKangFanghigh-dose+CTX group. We Observed changes in the living conditions of mice; We alculatedthe average tumor mass of mice and the inhibition rate and the immune organs of mice spleen,thymus, and mice in each group; We observed the tumor cell ultrastructure by electronmicroscope and observed the morphological changes of tumor cells in mice in lightmicroscope, We also used immunohistochemical method to detect the murine Lewis lungtumor cells mtp53, Bcl-2gene expression.Results1、The general state of mice: tumor-bearing mice model thin, clusters, coat drying; CTXmice coat dull, apathetic, slow movement, a small amount of food; Hui medicine AiKangFangin mice fur relatively smooth, the general mental state, exercise a little slow; Hui medicineAiKangFang the low-dose with a simple+CTX group were similar Icahn side; Hui medicineAiKangFang of high-dose+CTX group: fur smooth, good spirits, flexible movement weighthad increased.2、The inhibition rate: After treatment, mice with Lewis lung tumor in each group werereduced to varying degrees: Hui medicine AiKangFang square group of high drug doseinhibition rate of the best+CTX group (71%), pure Hui medicine AiKangFang,39.62%, CTX group of34.29%. CTX group can be seen around the cancer nests small amount of fibroustissue, focal necrosis, and inflammatory cell infiltration can be seen; back side drug Icahngroup see a small amount of apoptosis, apoptotic cells become smaller, rounded, nuclearcondensation, cytoplasmic concentration, staining deepened, no inflammation aroundapoptotic cells; Hui medicine AiKangFang low-dose group+CTX group health side much thesame microscope, but more can be seen a large amount of apoptotic cells and necrosis.Electron microscopy: tumor model group were round or polygonal tumor cells, many cellsurface microvilli, irregular nuclear shape, a notch, prominent nucleoli, multiple nucleoli, noswelling of mitochondria; Hui medicine AiKangFang side group smaller volume of tumorcells, cell swelling, vacuolization phenomenon occurs, mild nuclear pyknosis, gathered in thenuclear membrane; CTX tumor cell nucleus fragmentation, necrosis was the performance;Hui medicine AiKangFang of high-dose+CTX group nuclei of tumor cells shrinkage,chromatin condensation, gathered in the nuclear membrane, the cytoplasm can be seen morelysosomes, mitochondria swelling, change into a round, Jing fewer shorter or even disappear,in extreme swelling into a vacuole-like structure when and structure of an apoptoticcharacteristic of apoptotic bodies.5、Immunohistochemistry: mutant p53was mainly localized in the nucleus, there aresome lightly stained cytoplasm, Bcl-2located in the cytoplasm. After treatment p53, Bcl-2expression were decreased compared with the control group Hui medicine AiKangFang sideeffects of high dose+CTX group was significantly (P <0.01). Conclusions1、Hui medicine AiKangFang significantly improve the quality of life of lung cancer ofmice, high-lose was the best.2、Hui medicine AiKangFang can inhibit Lewis lung carcinoma cell growth;3、Hui medicine AiKangFang with chemotherapy reduced to some extent, damage ofchemotherapy on the role of immune organs;4、Hui medicine AiKangFang effects may be synergistic anti-cancer chemotherapy byreducing mutant p53and Bcl-2expression achieved, the promotion of tumor cell necrosis andapoptosis.5、The study of Hui medicine AiKangFang could become the basis for further study of theHui medicine treatment cancer.
