Objective:To detect the expressions of nuclear factor kappa B(NF-κB)and aquaporin-3(AQP-3)in human degenerated lumbar discs by immunohistochemistry, to explore the roles ofNF-κB and AQP-3in the process of intervertebral disc degeneration. Moreover, weexplore the relevance of expression of NF-κB and AQP-3in the process of intervertebraldisc degeneration to provide a new strategy for the research and treatment ofintervertebral disc degeneration.Methods:1. Selection of specimen: we obtained degenerated lumbar intervertebal disc tissuesform30patients with lumbar disc herniation by surgical operation in our departmentfrom June2012to February2013. We observed the intervertebral disc degeneration ofpreoperative by MRI, in accordance with the Videman grading scoring criteria weredivided into three groups: A group score1to3points is mild intervertebral discdegeneration group; B group score4to6points is moderate intervertebral discdegeneration group; C group score7to9points is severe intervertebral disc degenerationgroup.2. Detection of NF-κB and AQP-3: we detected the expressions of NF-κB andAQP-3in degenerated lumbar discs by immunohistochemistry. And to explore NF-κBand AQP-3interaction in the process of disc degeneration.3. Statistical analysis: weanalyzed the expression of NF-κB and AQP-3by Image proplus6.0image analysissoftware. The data was analyzed by SPSS17.0statistical software. Results:The expression of NF-κB was gradually increased with the aggravation ofintervertebral disc degeneration, while AQP-3was gradually reduced(P<0.01). Moreover,the expression of NF-κB and AQP-3was negatively correlated in the process(r=-0.805,P<0.01).Conclusions:1.Both NF–κB and AQP-3were detected in the degenerated intervertebral disc.2.There exist clear correlation between expression of NF–κB and AQP-3.The activationof NF–κB signaling pathway may be one of the important molecular mechanisms thatlead to the expression of AQP-3weakened in degenerated intervertebral disc.
