Objective:To research the expression and its relationships between NF-kB and MMP-9in the disc degeneration in different time points, and to dicover their functions and significance.Methods:To establish animal model of disc degeneration by puncturing intervertebral disc fiber ring along with the road of the right retroperitoneal operatioin. Disc specimens were taken out in different time points postoperation to observe the changes of general structure and the changes in microscopic structure by slicing and HE dyeing, the double mark immunohistochemical slices of NF-kB and MMP-9Using MaXvisionTM double mark immunohistochemical assay were manufactured to observe the expression of NF-kB and MMP-9.Results:1. The expression of MMP-9in those cells NF-kB positive expressed and its matrix around was that:there was little quantity of expression in4weeks; expression increased compare to other regional at8weeks:At12weeks, it is higher of the MMP-9expression compare to other regional.but it is down compare with after8weeks; There were no obvious differenc of the MMP-9expression compare to other regional after24weeks.2. The expression of NF-kB in those cells MMP-9positive expressed was that: there was little quantity of expression in4weeks; expression increased compare to other regional at8weeks; There were no obvious differenc of the NF-kB expression compare to other regional in MMP-9negative expressed cells after12weeks and24weeks.3.The cell double expression of NF-kB and MMP-9in the cartilage endplate was that:there was little quantity of expression in4weeks; there was highter expression in8weeks; then it descent in12weeks compare with8weeks, and there was no expression in24weeks.Conclusion:1. There was a trend of the double expression cells of NF-kB and MMP-9that increased in the early stage and then gradual decrease in the disc degeneration cartilage endplate. and the heightest expression appeared in8weeks.2. The expression of MMP-9may be up-regulated by NF-kB in the early stage of the disc degeneration.3. The high expression of MMP-9may be one of the main causes of the decrese of NF-kB in the disc degeneration, it may have a negative feedback adjustment effect to NF-kB.
