Objective:To investigate the expression of nuclear factor E2 related factor 2(Nrf2) and the content of reactive oxygen species(ROS) in human intervertebral disc and explore their possible relationship and role in the different degree degeneration. Methods:The intervertebral disc tissues were collected from 40 patients who performed spinal surgeries at the First affiliated Hospital of University of South China from February to November in 2015. According to MRI Videman grading scoring criteria, the samples were divided into four groups. Group A : 0 point, was deemed as normal control group(samples from patients with thoracolumbar acute fractures); Group B: 1 to 3 points, was deemed as mild degeneration; Group C: 4 to 6 points, was deemed as moderate degeneration; Group D: 7 to 9 points, was deemed as severe degeneration. Immunohistochemical method and fluorescent probe were used to detect the expression of Nrf2 and the content of ROS respectively. The Image pro-plus 6.0 software was performed to analysis the expression of Nrf2, and the SPSS18.0 statistical software was applied to analysis the data. Results:The experimental results show that the mean option density of Nrf2 in group A,B,C,D were 0.0749±0.0142, 0.0533±0.0064, 0.0344±0.0106, 0.0162±0.0054 respectively, and the content of ROS in group A,B,C,D were 181.558±13.148,201.154±17.510,220.437±19.023,237.467±23.704 respectively. The content of ROS was gradually increased with the aggravation of intervertebral disc degeneration(p<0.05),while the expression of Nrf2 was gradually reduced(p<0.05). There was a negative correlation between ROS and Nrf2 in the process of intervertebral disc degeneration(r=-0.733, p<0.05). Conclusions:1. In the human intervertebral disc tissue, with the aggravation of intervertebral disc degeneration, the expression of Nrf2 decreases and ROS content increases, there is a negative correlation between them. 2. Nrf2 may participate in the intervertebral disc degeneration.
