BackgroundCyr61is an immediate early gene and an important regulator of cell matrixstromal. Cyr61has obvious mitogenic and chemotactic activity, involved in the regulation of cell proliferation, transfer, adhesion and the formation of blood vessels. Cyr61express in a variety of biological functions in tumor, and Cyr61expression levels and functions are different in different tumor tissues and cells. However, the exact relationship between Cyr61and the tumor and its mechanism is not clear.In this study, we analysed Cyr61expression and significance in Human extrahepatic biliary system cancer, we observed the proliferation. This study established the foundation for further studies on the function and mechanism of Cyr61in tumorigenesis, and provided a new tumor marker and a gene therapy target for tumor diagnosis and therapy.ObjectiveTo investigate the expression of Cyr61in Human extrahepatic biliary system cancer and normal, and to explore the relationship of Cyr61and Human extrahepatic biliary system cancer.Methods(1) Using immunohistochemistry detects the expression of Cyr61in30cases of Human extrahepatic biliary system cancer and normal tissues.(2)Using real-time RT-PCR detects the expression of Cyr61mRNA in30cases of Human extrahepatic biliary system cancer and normal tissues.ResultsResults of immunohistochemistry:the positive rates of the expression of Cyr61in cancer tissue and normal tissue were100.0%and13.3%respectively. The difference between the two group was significant(P<0.001).(2)Results of real-time RT-PCR:The median quantity of Cyr61mRNA in Human extrahepatic biliary system cancer tissue was3.757(2-△CT x100000),while the median quantity of Cyr61mRNA in normal tissue was5.240(2-△CT x100000), the difference between the two group was significant(P<0.05).Conclusion1. Cyr61is expressed both in Human extrahepatic biliary system cancer tissue and normal tissue.2. The expression of Cyr61is up-regulated in Human extrahepatic bleary system cancer.3. The expression of Cyr61was no difference in the gallbladder and extrahepatic cholangiocarcinoma.
