Objective Through the observation of p15and HSP70gene expression in extrahepatic cholangiocarcinoma, carcinoma of the extrahepatic cholangiocarcinoma tissuesand normal bile duct tissues, and explore their relationship and clinical significance.In order to provide experimental evidence for the clinical treatment of extrahepaticcholangiocarcinoma.Methods By immunohistochemistry(SP) detection of p15and HSP70gene in humanextrahepatic cholangiocarcinoma, extrahepatic cholangiocarcinoma tissue and normal bileduct tissue expression levels, western blotting detection in normal extrahepatic bile ductand extrahepatic cholangiocarcinoma, p15and HSP70protein levels in the organization.Results (1) Through the experiment, found in this study,40cases of extrahepaticcholangiocarcinoma tissue p15protein positive expression in9cases, positive rate was22.5%, the weakly positive in6(15%), moderately positive in3(7.5%), without a strongpositive expression.40cases of bile duct carcinoma tissue adjacent to carcinoma, thepositive expression of27cases, the positive rate (67.5%), including weakly positive in2cases (5%), moderately positive in16cases (40%), strong positive9cases (22.5%). In20cases of normal bile duct tissues and14cases of positive expression of positive rate (70%),including weakly positive in2cases (10%), moderately positive in5cases (25%), strongpositive in7cases (35%). Differences between groups, through the analysis of the two testclearly shows that, the final result of carcinoma tissues and adjacent tissues and normaltissues of p15expression were statistically significant (P <0.05), p15protein expression islower than the normal bile duct tissue and tissue adjacent to carcinoma of bile ductcarcinoma (P <0.05), and the tissue adjacent to carcinoma and normal bile duct tissue p15expression of no statistical difference (P>0.05). P15protein positive dyeing, tan thannuclei cytoplasm.(2) In40cases of extrahepatic cholangiocarcinoma tissue HSP70proteinexpression in30cases, the positive rate was75%, in which weak positive6cases (15%), moderate positive10cases (25%),14cases had strong positive (35%).40cases ofextrahepatic cholangiocarcinoma tissue adjacent to carcinoma, the positive expression of13patients, the positive rate of32.5%, among them4cases were weakly positiveexpression, moderate positive expression of6cases,3were strong positive expression. In20cases of normal bile duct group,13cases of negative, positive in7cases, the positiverate of35%, the weakly positive expression in2cases,4cases were moderate positiveexpression, strong positive expression in1case. Differences between groups, through theanalysis of the two test clearly shows that, the final result of extrahepaticcholangiocarcinoma tissues and adjacent tissues and normal tissues of HSP70expressionwere statistically significant (P <0.05), HSP70protein expression in extrahepaticcholangiocarcinoma tissue was higher than normal bile duct tissue and tissue adjacent tocarcinoma of bile duct carcinoma (P <0.05). And tissue adjacent to carcinoma and normalbile duct tissues of HSP70expression no statistical difference (P>0.05). HSP70proteinpositive dyeing dark yellow, deep than nucleus to cytoplasm staining.(3) the p15positiveexpression rate is associated with tumor grade (P <0.05). HSP70positive expression rateis associated with tumor grade (P <0.05).(4) the p15protein and HSP70protein ofWestern blotting tests showed:40cases of extrahepatic cholangiocarcinoma tissue p15proteins were lower expression, and significantly lower than the normal bile duct tissueand tissue adjacent to carcinoma; In40cases of extrahepatic cholangiocarcinoma tissue,HSP70protein expression were high status, significantly higher than the normal bile ducttissue and tissue adjacent to carcinoma. In p15positive expression of extrahepatic bileduct carcinoma tissue, HSP70expression rate is low, the Pearson related analysis andcalculation, the results show that the extrahepatic cholangiocarcinoma tissue of p15expression in negative correlation with the expression of HSP70, with statisticalsignificance.Conclusion (1) The p15proteins show lower expression in the extrahepaticcholangiocarcinoma tissue, HSP70protein was highly expressed in the extrahepaticcholangiocarcinoma tissue;(2) The p15and abnormal expression of HSP70associated withthe degree of tumor differentiation of extrahepatic cholangiocarcinoma;(3) The p15and theexpression of HSP70in the extrahepatic cholangiocarcinoma showed a negative correlation..
