The Study Of Susceptibility Genes Of Type 2 Diabetes Mellitus In Northern Chinese

Type 2 diabetes mellitus (T2DM) is one of the complex syndromes which caused by both of hereditary factors and environmental factors, and has strong genetic susceptibility. In this study, we selected eight susceptibility genes with polymorphisms as candidates of T2DM, which had not been studied in Northern Chinese. Case-Control Method and PCR-RFLP technique were used in order to study the associations between these susceptibility genes and T2DM. 295 T2DM patients (153 males, 142 females ) and 319 normal controls (181 males, 138 females ) from Beijing and Haerbin were investigated.The eight susceptibility genes' polymorphisms included the Ser 20 Gly ( A-G ) polymorphism in islet amyloid polypeptide (IAPP) gene, the C-T polymorphism in intron 14 of glycogen synthase (GYS1) gene, the 2549 C-A polymorphism in the promoter of leptin gene, the Gln 223 Arg ( 668 A-G ) polymorphism in leptin receptor (Lper) gene, the -3826 A-G polymorphism in uncoupling protein ( UCP ) gene, the T - C polymorphism in intron 6 of lipoprotein lipase (LPL) gene, the Ala 54 Thr ( G-A) m in intestinal fatty acid binding protein (FABP2) gene and the Gly 15 Gly ( T-G ) polymorphism in adiponectin (ADPN) gene. In addition, we screened 9 exons of paired box 4 (PAX4) gene to seekpolymorphisms associated with T2DM.The conclusions were as follows: (1) No polymorphism associated with T2DM was found on PAX4 gene, which suggested it wasn't susceptibility gene of T2DM. (2) The intron 14 C-T polymorphisms on GYS1 gene showed significant relative with T2DM. Significant differences were found in the frequencies of the genotype CC and CT, allele C and T. (3) The 2549 C-A polymorphism in the promoter of leptin gene showed significant relative with T2DM. Genotype CC had positive relation with the risk of T2DM. (4) The G1n 223 Arg polymorphism in Lper gene were associated with T2DM. Genotype AG and allele A had positive relation with the risk of T2DM, genotype GG and allele G had nigative relation with the risk of T2DM. (5)The Ser 20 Gly (A-G ) polymorphism in IAPP gene, the -3826 A - G polymorphism in UCP gene, the T - C polymorphism in intron 6 of LPL gene, the Ala 54 Thr ( G-A ) polymorphism in FABP2 gene and the Gly 15 Gly ( T-G) polymorphism in ADPN gene were not associated significantly with T2DM in Northern Chinese. (6) The degree of correlation between the polymorphisms on these genes and T2DM showed certain variance under different backgrounds.
