Backgrounds:Melanosis coli(melanosis coli, MC) is a kind of Gastrointestinal melanosis, whichreveals pigmentation in Colonic mucosa. Research shows that melanosis coli is relation toAnthraquinones cathartic. Laxative is a main treatment used by traditional Chinesemedicine. Moreover, a lot of abuse drug reducing weight in traditional Chinese medicinewhich contain laxative result in the increase of Melanosis coli year by year. Most scholarsbelieve that anthraquinone laxatives and anthraquinone compounds lead to Melanosis coliby Animal experiments and the detection of endoscopic colon mucosa, which Plays animportant role in the development of Melanosis coli and become pathopoiesia crim.dose-effect relationship is a key factor when Anthraquinones laxatives and anthraquinonecompounds result in MC, which Remains to be further researched. The topic of Ourresearch is to develop an HPLC method for determination of the pharmacokinetics ofemodin in anthraquinone of herbs in guinea pig.Meantime, to detect the pathologicalchanges of Melanosis Coli with HE staining and transmission electron microscope, and study the expression of Bcl-2and Bax in the colon of guinea pig; also apply TUNEL todetermination of apoptosis of colon muscosal epithelial cells.Objective:To detect the pharmacokinetics of emodin in anthraquinone of herbs in guinea pig,and investigates effects of apoptosis of colon muscosal epithelial cells on melanosis colifor its relationship to clinical pathological characteristics of MC, therefor explore theconnection of melanosis coli resulted from anthraquinone of herbs.Methods:To develop an HPLC method for determination of the pharmacokinetics of emodin inanthraquinone of herbs in guinea pig. Using HE staining and transmission electronmicroscope to detect the pathological changes of Melanosis Coli, and studying theexpression of Bcl-2and Bax in the colon of guinea pig; also apply TUNEL todetermination of apoptosis of colon muscosal epithelial cells.Results:The intra-day and inter-day RSD was1.76%and2.43%, and the average recoveryrate was92.05-98.55%. Emodin were completely separated and no interference wasfound,also showed good linearity of the concentrations from4to28ug/ml(r=0.9996).Excepted for the normal controls group, the inflammatory changes were discovered inevery group such as neutrophil infiltration and different degrees of epithelial cell sheddingby HE staining. the inflammatory changes of lymphocyte and leukocyte infiltrationfollowed mitochondrial swelling were observed by transmission electron microscope inthe slower Radix Ophiopogonis group and the slower Rhubarb group. Scattered on thelipofuscin was discovered in in the higher Radix Ophiopogonis group, Apoptosis andnecrosis of cells were found in the higher Rhubarb group. The positive rates of Baxexpressions was significantly increased in the higher Rhubarb group compared Withnormal controls (p <0.01), Also, the expression level of Bcl-2was significantly higher inthe slower Radix Ophiopogonis than the normal controls, The apoptotic index of controlgroup was significantly slower than the higher Rhubarb group.Conclusion: The content of emodin in anthraquinone evacuant was obviously higher than RadixOphiopogonis in anthraquinone compound. The apoptosis of colon muscosal epithelialcells was likely one of the factors and mechanisms caused by melanosis coli, and waspositive correlation compared with the level of melanosis coli.
