ObjectivesThe aim of this study is to investigate the protective effect and mechanism of edaravoneinjection on hepatic injury in rats with severe acute pancreatitis by animal experiments, and toobserve the changes in serological markers like AMY, AST, ALT, TNF-, IL-6, SOD and MDAduring the courses of severe acute pancreatitis.Methods72Male Sprague-Dawley (SD) rats were randomly divided into3groups with24rats in eachgroup-Sham operation group (SO Group); Severe acute pancreatitis model group (SAP Group);Edaravone group (EDA Group).1ml/kg of4%sodium taurocholate solution was injected byretrograde puncture of pancreatic duct through duodenum in order to set up SAP model.3mg/kgof edaravone was injected intraperitoneally in EDA Group. Same volume of normal saline wasinjected intraperitoneally in SAP Group. Open incision in abdomen was made after anesthesia inSO Group, then turning the duodenum and pancreas, and abdominal injection of an equal volumeof saline. Normal saline (30ml/kg) was injected subcutaneously in all groups to compensate thewater loss during modeling. Blood samples and specimens from pancreas and liver were takenfor tests at4,8,12hours (n=8in each time point) after modeling. Test indices including serumamylase (AMY), serum alanine aminotransferase (ALT), serum aspartate aminotransferase(AST), serum tumor necrosis factor-(TNF-), interleukin-6(IL-6), serum superoxidesuperoxide dismutase (SOD), serum malondialdehyde (MDA), and pathological score ofpancreas and liver tissues. All the data collected was analyzed by SPSS13.0for windowsstatistical software.ResultsSerum markers and the pancreas, hepatic pathology scores were no significant changes in SOgroup, the3groups at each time point compared to each other were not statistically different (P>0.05). Serum AMY, ALT, AST, TNF-, IL-6and MDA levels of SAP Group in the3time pointswere higher than that in SO Group. SOD was lower compared with SO Group (P<0.05).Pancreas and liver pathological score was higher than that in SO Group (P<0.05). Indices inEDA Group and SAP group improved with the time at the3time points (P<0.05). Pancreas and liver pathological score was lower than that in SAP Group (P<0.05).Conclusions1.4%sodium taurocholate injection by retrograde puncture of pancreatic duct throughduodenum is an easy way to set up SAP and hepatic injury model. It is simple, reliable,reproducible, and an ideal SAP modeling.2. TNF-, IL-6, MDA and SOD is an importantparameter of inflammatory reactions in SAP and it is an important factor of SAP with hepaticinjury.3. Edaravone can lower the degree of hepatic injury in SAP. Its mechanism may be relatedto the inhibition of serum inflammatory mediators such as TNF-, IL-6generation, andclearance of excess free radicals in the body and reducing the loss of liver cells.
