The bio-effect of electric field treatment seed have been studied for a long time, however, it’s still not developed widely and utilizated effectively. One of the important reason is that the mechanism of electric field treatment seed bio-effect is unclear.Based on the previous research on the drought resistance of plant seeds after electric field treatment, using the mRNA differential display technique at the plant DNA transcription level, this report try to explain electric treatment plant seeds macro bio-effects linked to micro mechanism. Caragana korshinskii seeds and young leaves were chosen respectively as the research object and materials, we used total RNA which was extracted from young leaves as the template, then adopted3kinds of anchor primers and26kinds of random primer combination for DDRT PCR. After gel electrophoresis on modified PAGE and staining by silver nitrate, we got41Significant difference segments whose length from200bp to750bp.After cloning and sequencing, we designed specific primers based on the results of the sequencing to carry out RT-PCR verifying. Finally, we got three positive difference segments and their length is534bp,308bp and472bp respectively. After BLAST compare the sequence of the three fragments, we obtain their homologous proteins which they may transcribe and translated. Through discussing the function of their homologous proteins, we made analysis for the impact on the growth of Caragana korshinskii.
