Lilium concolor and Lilium davidii are both native wild lilies, originating from China. And they are kinds of perennial bulbous flower belong to Liliaceae, Lilium. In this experiment, We used the plantlets scales and leaves of Lilium concolor and the lily bulb scales of Lilium davidii as explants, which were cultured in respective MS medium supplemented different hormone combinations. Then the culture ways of the two different kinds of lilies were explored, such as calli induction, adventitious buds differentiation, bud proliferation and rooting,established a complete technological system of tissue culture and rapid propagation.Results for tissue culture and rapid propagation of Lilium concolor:(1) Calli induction: the seedlings scales were the most suitable explants, calli induction rate was higher and the growth was better. The most suitable culture medium for calli induction was MS+6-BA (1.5mg/L)+NAA (1.Omg/L), and the induction rate was as high as85.33%.(2) The differentiation of calli and bulblet regeneration: the optimal medium for calli differentiation was MS+6-BA (1.0mg/L)+NAA (0.5mg/L), the rate of differentiation was66.04%, the average amount was5.38, and the majority of the buds had developed into little bulblet.(3) The enlargement of the bulblet: the best medium was MS+6-BA (1.0mg/L)+NAA (0.5mg/L), the average bulblet weighed0.25g, and the weight coefficient was11.2.(4) The rooting culture: for mono-factor-influence such as MS, sucrose, AC, the results suggested1/2MS,70g/L sucrose and0.3g/L activated carbon each effect best on the bulb rooting, their average root amount was3.10,2.83and3.87respectively. But the concentration of sucrose had no significant effect on rooting, took the cost of sucrose and its effect on rooting into consideration, the best choice was30g/L, and its average root amount is2.63.Results for tissue culture and rapid propagation of Lilium davidii:(1)Primary culture: for the explants of bulb scales, the best disinfection scheme was disinfection with alcohol for90s, then0.15%HgCl2for12min, its contamination rate was9.72%and the necrosis rate was only16.67%; the differentiation ability of outer layer scales was the strongest,their induction rate was73.67%; the best medium for adventitious bud induction is MS+6-BA (1.0mg/L)+TDZ (0.2mg/L), the induction rate was as high as86%, and the average sprouting numbers performed well, is4.3.(2)Bulblet induction and proliferation: the optimal medium was MS+6-BA (1.5mg/L)+NAA (0.5mg/L), bulblet proliferation coefficient was as high as3.93.(3)Rooting culture:the best rooting cultivation medium was MS+IBA (0.2mg/L), the average root number was10.13, and the bulb was stouter,the root stronger.
