Lilium speciosum Thunb. var. gloriosoides Baker belongs to Sect. Sinomartagon of Liliaceae, a variant of Lilium speciosum. The species is distributed in Anhui, Jiangxi, Zhejiang, Hunan and Jiangxi. And grew down there in the damp undergrowth and grass of hillside. And the fields about370~900m above sea level.The test material from the Shanshan forest farm in the region of Wannan Mountains in Anhui. The forest is located in Xiaoken and tangji. It has a subtropical monsoon climate zone. Their geographical position is (30°19’-30°27’N,117°39’~117°48’E). Zonal vegetation is evergreen broad-leaved forest.In the paper, we studied the culture in vitro of Lilium speciosum Thunb. and explored on the cultivation technique. We have established the Lilium speciosum Thunb. breeding system for the protection and development of Lilium speciosum Thunb.. The results provide the theoretical basis for in-depth study and development of Lilium speciosum Thunb.. The main results are as follows:1.The more suitable sterilization treatments for squamas of Lilium speciosum Thunb. was as70%C2H5OH30s+0.01%HgCl220min.2. The best explants is healthy and no lesion in the middle scales.3.Scales placed:The inside(adaxial) facing up on the medium.4.The effeets on buds propagation of three basic factors sucrose>NAA>6-BA. The optimal medium for callus is MS+1.5mg/L6-BA+0.5mg/L NAA+30g/L Sucrose. The optimal medium for buds propagation is MS+1.5mg/L6-BA+1.0mg/L NAA+30g/L Sucrose 5.The optimal subculture medium for callus is MS+1.5mg/L6-BA+0.5mg/L NAA+30g/L sucrose; The optimal subculture medium for buds propagation is MS+1.5mg/L6-BA+1.0mg/L NAA+30g/L Sucrose6.The optimal Rooting medium for buds propagation is1/4MS+1.0mg/LNAA+10g/L Sucrose...
