| Lilium davidii var. unicolor scales were used as material in this study,and the reproduction effect of the experiment was optimized according to the single-factor and two-factor randomized blocks under different temperature and light conditions,and the best aerobic culture environment was screened out.The relationship between the carbohydrate metabolism law of different scale levels and scale weights in the cultivating process and the formation of small bulbs under optimal environmental factors was studied,In order to solve the problems of low reproductive efficiency of Lilium davidii var.unicolor in the air culture breeding process,low bulb quality and long reproduction period,and high scale infection rate,this study proposed a more reasonable scale breeding technology system.The main findings are as follows:(1)The influence of aeroculture environment on the effect of accelerated culture.Temperature single-factor aerobic culture test showed that the scale differentiation rate was the highest,the small bulb grain weight was the largest,and the number of roots was the largest when treated at 25°C at various aerocultivating temperatures(15℃、20℃、25℃、30℃);The rooting rate of small bulbs is the highest;the differentiation number of small bulbs is the highest under the treatment of 20℃ and25℃,and there is no difference between the two,reaching 146.67 grains/100 pieces and 146.48 grains/100 pieces respectively.Under various light intensities(0Lx、500Lx、1000Lx、1500Lx、2000Lx),the comprehensive effect of cultivating under500 Lx treatment is the best,with the highest differentiation rate of 95.33%,and the differentiation number of small bulbs up to 278.00 grains/100 pieces.At the same time,the rooting rate is the highest and the number of roots is the highest.Principal component analysis was carried out on the 6 observation indexes of the two-factor randomized block test of temperature and light,which were divided into 5 principal components.The cumulative variance contribution rate of the first three principal components reached 93.99%,and the eigenvalue of the first principal component was3.988.It mainly reflects the differentiation rate of scales and the number of small bulbs and the differentiation of small bulb roots;the second principal component feature value is 1.1772,which mainly reflects the weight of small bulbs;the third principal component feature value is 0.4744,which mainly reflects the small bulb grading.The comprehensive evaluation results based on the 6 observations of each combination of the two-factor test of temperature and light show that the effect of cultivating scales is the best under the environment of 25℃+500LX,which can be used for large-scale reproduction of scales.(2)The effect of scale layer and scale weight on the reproduction effect of aeroculture.Divide the scales into three layers: outer layer(1-2 layers),middle layer(3-4 layers)and inner layer(5-6 layers).Cultivate them at 25℃+500LX respectively.The middle layer has the highest differentiation rate(93.00).%),the number of small bulbs is the most differentiated(213.67 grains/100 scales),and more than 91% of the scales can differentiate into 1-4 small bulbs.According to the weight of the scales,the scales are divided into four levels,of which the third-level scales have the best effect.The scale differentiation rate reached 93.25%,and the number of small bulb differentiation was 144.50/100 pieces.The larger the scale quality,the lower the scale differentiation rate,the lower the rooting rate,and the fewer roots.(3)The influence of light intensity on the carbohydrates of scales and small bulbs during the aeroculture process.The carbohydrate content in scales and small bulbs changes with the change of light intensity.All light intensities at the optimal incubation temperature(25℃)cause the starch content of scales to decrease,and the soluble sugars increase slowly.Among them,500 LX starch is the most degraded.Fast,the starch content in small bulbs is relatively stable,and the change of soluble sugar and reducing sugar is not significantly affected by light intensity.The carbohydrate changes of the scales during the incubation process once again confirmed that 25℃and 500 Lx are the best incubation environment. |
