Ketamine, xylazine and midazolam are three kinds of drugs commonly used in veterinary clinics, in particular, more commonly used for small animal clinical anesthesia. But, one anesthtic used alone is often difficult to achieve the desired anesthetic state and has obvious side effect. Shumianning combination which containe ketamine, xylazine and midazolam, is developed by the Small Animal Disease Laboratory, Nanjing Agricultural University. The aim of this experiment was to evaluate the effects of Shumianning on anesthetic quality, cardiopulmonary system and the function of the liver and kidney in puppies. Comparison of the anesthetic effect of both Shumianning and Telazol and evaluation of their problems were made after intramuscular injection of them in puppies, The anesthetic quality and function of liver and kidney were further evaluated when Shumianning combination was injected intravenously with clinical surgival patients in order to provide data and reference for clinical application of it.Eight healthy puppies were intravenous injected with Shumianning combination (0.01mL·kg-1IV) within60s without premedication. The onset of anesthesia was rapid in puppies and the initiation, maintenance and recovery time was21.4±2.1s,28.0±1.1min,18.0±1.4min after administration of Shumianning; After anesthesia blood pressure of puppies increased first, then decreased slowly but remained within the normal physiological range; During maintenance of anesthesia the HR and RT decreased slowly (P<0.05); RR decreased significantly at5min after anesthesia and then increased to normal range; SpO2was lower than the normal physiologic range at1min but increased to normal range after5min. All the biochemical parameters were in the normal physiologic range. We can conclude that no obvious influence of Shumianning was observed in the function of liver and kidney in young dogs was small, No adverse reaction such as vomiting and profuse salivation was found in the period of anesthesia.Healthy puppies were divided into Shumianning anesthesia group (8puppies) and Telazol anesthesia group (8puppies). Both Shumianning and Telazol were administered by intramuscular injection, all the puppies were premedicated with atropine. There was no significant difference between Shumianning and Telazol group in anesthetic initiation time, but Telazol group had longer maintenance and recovery time than Shumianning group. Effects of sedation and muscle relaxation were better in Shumianning group than in Talazol group, which is significant difference between twi groups (P<0.01). There was no significant difference in pain scores in the two groups. Puppies could be successful intubated endotracheally and didn’t have flatulence and excessive salivation in Shumianning group. But they could be difficult to be intubated, represented the reflexe of swallowing, laryngeal, palpebral and flatulence and excessive salivation for some dogs in Telazol group. There was no significant difference in pain scores in the two groups (P>0.05). RR, RT, SpO2and PETCO2were not significantly changed (P>0.05) between Shumianning and Telazol group. In Shumianning group, the pup’s HR and blood pressure increased first and then decreased slowly but remained within the normal physiological range. Tachycardia happened in Telazol group.Shumianning injection was administered in sixty dogs which were operative in clinic. The results showed that The time of the operation (T3) and awaking (T4), the adding times(N) of Shumianning, the consumption of Shumianning (SUM) were dependent on the operation type and the body size. The maintenance time was23.8±8.1min, the awaking time was32.2±12.9min, the recovery quality score was3.8±0.5. Anesthesia quality was excellent (51/60,85.0%), good (7/60,11.7%), and fair (2/60,3.3%). Recovery score was3.8±0.5. There is sable was stable, no ataxia in recovery stage and postoperative recovery was good. GLU was significantly higher than that of normal physiological levels (P<0.01). From the view of anesthesia quality for operation in clinical dogs, Shumianning injection is suitable to the clinical anesthesia of dogs. It is cherecteristic of satifactory anesthetic effect and smooth recovery during the mantanaince anesthesia and recovery tages.
