Aucubin is a kind of iridoid glycoside with extensive biological activities, which contains in eucommiaceae, plantago, scrophulariaceae, caprifoliaceae and other plant. The studies have shown that the eucommia barks, leafs and seeds all contains high amount of aucubin, the concentration of Aucubin in the eucommia leafs is relatively higher. The research and utilization of eucommia were mainly focused on barks and seeds of eucommia, but seldom on leaves. With further research on aucubin, people should pay more attention on extracting aucubin from the eucommia leaves, instead of from the eucommia bark and seeds.In this paper, eucommia leaf and the aucubin containing as object, aucubin was determined by UV spectrophotometry and HPLC method, through the study of the extraction solvent, extraction time, extraction temperature, extraction conditions. The S-8macroporous resin for separation purpose of extraction and purificationfrom the aucubin in solution was established; the n-butanol crystallization of aucubin verification work. Finally, a set of industrialization process to obtain aucubin from eucommia leaf.1. By using the spectrophotometer was used to examine the linear relationship between the different concentrations of aucubin methanol solution and the absorbance, and investigated the relationship between the stability of aucubin. The results show that: aucubin stability with temperature, pH, light has a certain relationship. When the temperature exceeds60℃easy decomposition. In acid and alkaline solutions are not stable, need to control on the condition of pH6is the most stable.2. Based on literature and the experiment method was established fordetermination of aucubin in sample solution by HPLC, Liquid phase chromatographic column is: Betasil C18(250mm×4.6mm,5℃m); the detection wavelength:210nm. Mobile phase: methanol water solution (10:90, V/V); Flow rate:1mL·min-1; column temperature:25℃. This method is stable and reliable, high sensitivity, suitable for other related experiments of aucubin was determined.3. In the extraction of single factor experiment based on orthogonal design factors of the experiment of aucubin, obtained using10times amount of80%ethanol extract of herb50min at50℃, extracting3times the extraction result is good.4. Through the study of literature and market investigation by the separation of aucubin in Eucommia ulmoides leaves with macroporous resin S-8, and the n-butanol crystallization method was verified. Results show that the use of S-8macroporous resin for separation, the aqueous solution of4.12mg/mL2BV/h of flow rate, per gram dry resin like20.60mg, theneluted with3BV40%ethanol3BV/h velocity, aucubin collect the eluent is concentrated by the purity of crude product of n-butanol crystallization of up to92.56%, the stability and reliability of the method.
