Eucommia ulmoides male flowers is a precious medicinal material resources inChinese and is rich in natural active substances. But the domestic and foreign relatedresearch are few. In this project, we have studied on the activity of Eucommiaulmoides male flowers adopting the methods thate solvent extraction, silica gel column,thin layer chromatography and biological activities. Furthermore, the Pharmacologicalmechanisms are discussed, and provide scientific basis for the rational utilization ofEucommia ulmoides male flowers resources.In this study, basising on single factor tests, the response surface methodologywas used to optimize extraction technological conditions for ethanol extracts, EtOACextracts, n-BuOH extracts, flavonoids. And the optimum extraction technologyparameters have been defined. The main active extracts of Eucommia ulmoides maleflowers were obtained through chromatography, sedative and hypnotic, antibacterialactivity and antitumor activity.The results showed that the optimum extraction technology parameters of ethanolextracts were: the extraction temperature62.81℃, the extraction time2.91h, theethanol concentration59.84%. The optimum extraction technology parameters offlavonoids for maximizing yield were treated for65℃,2.84h, ethanol concentration59.58%. The optimum extraction technology parameters of EtOAC extracts were: theextraction temperature50.50℃, the extracting time53.14h, ratio of liquid and rawmaterial10.12∶1. The optimum extraction technology parameters of n-BuOH extractswere treated for26h,52.98℃, ratio of liquid and raw material10.63∶1. The differentpolarity extracts of Eucommia ulmoides male flowers all have sedative and hypnotic,bacteriostatic. The active of EtOAC extracts, n-BuOH extracts, alkaloids hadmeaningful results. The water soluble alkaloids and fat-soluble alkaloids all hadsedative and hypnotic activities, and had synergy effects with pentobarbital sodium.The Water soluble alkaloids showed of anticonvulsions function. For seven of the bacterias, the Water soluble alkaloids and fat-soluble alkaloids only had an obviousinhibitory effect on Bacillus anthraci and the diameters of inhibition zone>15mm.The fat-soluble alkaloids showed the anti-tumor activities, could inhibit theproliferation of MDA-MB-231, IC50value were6.38mg/mL. The EtOAC extracts andn-BuOH extracts were separated, and the main active isolate were obtained. Samples1(methanol concentration10%) and sample2(methanol concentration20%) of theEtOAC extracts had certain antibacterial activity. It had an obvious inhibitory effect onStaphylococcus aureus, Bacillus anthraci, Bacilllus subtilis; and the diameters ofinhibition zone>15mm, also possessed of antibiotic effect on Escherichia coli,Aspergillusniger. Samples4(methanol concentration40%) of the n-BuOH extractshad an obvious inhibitory effect on Staphylococcus aureus and Bacillus anthraci; alsopossessed of antibiotic effect on Bacilllus subtilis, Escherichia coli, Aspergillusniger.The main active isolate were Samples1(methanol concentration10%) and Samples6(pure methanol with a few ammonia) of the n-BuOH extracts; their showed theantitumor activities, could inhibit the proliferation of MDA-MB-231and HepG2, IC50value were smaller than0.049g/mL. Their showed little effect on the normal cells.
