MARVELD1(MARVEL domain-containing1) is a member of MARVELdomain-containing proteins, which is highly conserved between mice and humans.Several studies have described that human MARVELD1is a tumor suppressor gene,regulating several genes’ expression and playing an important role in tumorigenesisand progression. Because of the highly conserved between mice and humans, weinvestigated the expression and distribution of mMARVELD1during embryogenesisThe expression and distribution of mMARVELD1during embryogenesis isinvestigated by in situ hybridization (ISH), quantitative real-time PCR (qRT-PCR)and immunohistochemistry (ICH). The results show that mMARVELD1wasprominently expressed in the nervous system including telencephalon,rhombencephalon and spinal cord during E9.5~E11.5mouse embryo. mMARVELD1and its protein was mainly expressed in the forebrain, midbrain, cerebellum, spinalcord, dorsal root ganglion, tongue, liver, heart and lung. Futhermore, weinvestigated the expression of some organs including brain, tongue, heart, lung andliver in E12.5, E15.5and E18.5mouse embryo. The results show that the expressionof mMARVELD1in E12.5is the highest, in E15.5is lower than that in E12.5, and inE18.5is the lowest. The expression of mMARVERD1in liver is the most significanthigh among other organs.Moreover, we have analysed2013bp DNA sequence in the upstream of thetranscription start site of mMARVELD1through Bioinfomatic methods. It turns outthat there are4GAGA sequences in this region. We tested the activity of promoterof mMARVELD1by dual luciferase reporter gene system. It shows that3GAGAelements which are close to the transcription start site show synergistic effect ofpromoting the transcription.In conclsion, mMAEVELD1and its protein were ubiquitously and highlyexpressed during mouse embryonic development, sugesting that it may play animportant role in the morphogenesis of multiple organs and tissues, especiallyassociated with brain and liver development at mouse embryonic stages.
