In order to study the morphological changes of cells, organs and tissues during continuous embryonic development in Kunbai strain mouse, the spatial and temporal distribution of early embryo,differentiation of germinal layer, and the development of system and organ after implantation were observed by using technology of paraffin section preparation and Hematoxylin & Eosin staining method.The expression and localization of CRB3 (one kind of polarity regulatory proteins) in different stages of mouse embryos and major organs and tissues of adult mouse were further studied to investigate the role of CRB3 on the polarity formation of embryonic development and epithelial cell of adult tissues in mouse by micromanipulating, RT-PCR and immunofluorescence staining technology.The results were as following:1. Morphological observations on mouse embryos showed that, the development of preimplantation embryos including fertilization and cleavage stages (including 2-cell stage, 3-cell stage, 4-cell stage, 8-cell stage, morula stage and blastocyst stage); through the formation and differentiation of three germinal layers based on gastrula after implantation, the establishment of basic body was completed, and then organ primordia formated and gradually developed completely. In this study, the morphological structure of mouse embryos at different stages were observed, and the systemic development atlas of mouse embryos were obtained. Our results could supply reference for the study of the migration of mouse embryonic cells, the changes in the relative position of organs within the embryo, and the continous study on embryonic development in mouse.2. CRB3 expressed both in oocytes and preimplantation embryos of mouse, and mainly distributed in the cytoplasm. The expression of CRB3 was higher in mature oocytes and zygotes compared to that in 2-cell embryos and 4-cell embryos, and gradually increased in 8-cell embryos, morula and blastocyst. CRB3 was also extensively expressed in embryo after implantation, including inner cell mass, trophoblast cells, yolk sac endoderm of 4.5d embryos, chorion, amnion, trophoblast, yolk sac endoderm, and embryonic ectoderm of 7.5d embryos, amnion of 8.5d embryos, semicircular canal epithelium, retina, lens bulbs, caudate lobe, and hepatic lobules of 13.5d embryo. CRB3 also expressed in the Uterus, stomach, kidney, liver, cerebellum, cerebrum and small intestine of adult mouse.The expression intensity of CRB3 protein in different organs and tissues, in sequences, were kidney, small intestine, stomach, uterus,liver, skeletal muscle, cerebellum and cerebrum.CRB3 was regularly distributed and located in the developmental mouse embryos, organs and tissues of mouse, suggesting that CRB3 protein played important roles in the establishment and maintenance of embryonic polarity and epithelial polarity of organs and tissues in mouse.
