GABA System Construction In The Brain And Suboesophageal Ganglion Of Drosophila Melanogaster

The experimental was conducted with our lab’s self-created technology platformof insect brain microscopic study, using Colophony-Paraffin,CP tissue embeddingdirectional microtomy technology combined with immunohistochemistry and rabbitanti-GABA polyclonal antibody, firstly labeld GABA immunoreactivity in the brainof Drosophila, and revealed the GABA-ergic system distribution in the brain andsuboesophageal ganglion of Drosophila melanogaster. The results shows as follows:strong GABA-like immunoreactivity appears in most functional brain structure ofDrosophila, such as ellipsoid of the central complex, optic lobes, olfactory lobe,mushroom body calyx and suboesophageal ganglion. Among these, ellipsoid is themost obvious staining structure and obvious stratification could be seen in the medullaof the optic lobe. According to cell position, size and morphological characteristics,these cells are divided into several groups. Seven groups appears in protocerebrum,respectively named SP, LH, IP1/IP2/IP3, SLP, VLP; Among these groups, IP2neurons axon fibers projecting to the ventral side and VLP neurons axon fibersprojecting to the superior protocerebrum could be clearly seen. Three immunoreactivecell groups appears in the olfactory lobe of the brain, respectively called AL1(outerlateral to olfactory lobe), AL2(inner lateral to olfactory lobe) and AL3(ventral toolfactory lobe).We can see obvious strong fibers from projection neurons projecting toprotocerebrum through antennocerebral tract. Seven immunoreactive groups appearsin the optic lobe: around medulla(M1-M3), LP1(dorsal to lobula plate),LP2(ventral tolobula), LP3(between lobula and protocerebrum), LP4(between lobula plate andprotocerebrum). Plenty of immunoreactive cells appears in suboesophageal ganglion,separately located ventral to suboesophageal ganglion SE1/SE4, outer lateral tosuboesophagead ganglion SE2, anddorsal to suboesophageal ganglion SE3. Besides,two big immunoreactive cells are arranged up and down called V1and V2This study fill in the blank of GABA-ergic system distribution pattern in thebrain of Drosophila melanogaster, our results coincide with gad1(glutamic aciddecarboxylase) in situ hybridization results under fluorescence before. But ourexperiment technique can get all the full fly brain serial sections, and can label manyweakly positive cells that other abroad laboratory can not label, what’s more,we canalso distinguish the detail position, number and shape of each cell group. In addition,according to the position of AL3group and their large projection fibers direction, weguess these cell groups belong to mACT projection neurons, and AL1group belong toantennal lobe local neurons when compared with previous study.
