The binding media is one of drawing material which has been widely found in ancient wall painting and cultural relics coloured drawing. They are employed to adjust the intersolubility of pigment and water, enhance the cohesion of pigment and wall, and make the paintings bright and remained so ever. However, the diseases which contained flaked, powdered, disrupted, mildewed and faded were all connected with binders. So the research of binding media becomes a hot topic in the preservation of cultural relics and restoring. The most common organic materials used as binding media are proteinaceous glues, gelatine and drying oils. The bases of gelatine is polysaccharides carbohydrates, it is refined from the secretion which were secreted by the stem and pole of plant. The proteinaceous glues are simmered from the skin, bone, horn and blood of animals. They are the most extensive application in ancient. Gas chromatography and liquid chromatography are commonly used for the detection of binding media. The theory is the proteins can be analyzed by its relative amount of amino acids. The content of amino acids in different glue are different, so we can determine every relative animal glue and sample’ glue, then confirm the result by comparison. The animal glue and sample must be hydrolyzed in order to release the amino acids, and these obtained amino acids are analyzed by separative techniques as gas chromatography or liquid chromatography. In this article based on the work of predecessors, the author consider that different proteins have different amino acid sequence, so we can determine the source of information of binders by amino acid sequence by Edaman degradation. Before exploring new methods, we can use the two classics liquid chromatography and Edman degradation obtained the source of information of the two binding medias of sample separately, then proving the usability of Edman degradation through comparison. The content is divided into five chapters.Introduction. Discussing the properties, source and determination methods of binding media, summarizing the advantages of previous work, putting forward the limitations of the study by consulting a large number of literature and then expounding the contents and innovations of this article.The studying of binding media by reversed phase High Performance Liquid Chromatography(HPLC) with precolumn phenylisothiocyanate derivatization(PITC). We determined the amino acid composition and proportion of3animal glues, egg glue and binding media by PITC-derivatization method, and confirmed the source of information of binding media is animal glue.The studying of binding media by reversed phase High Performance Liquid Chromatography(HPLC) with precolumn o-phtaladehyde(OPA)deivatization.We obtained the same conclusion in using OPA derivatization for the determination of the sample above.The preliminary exploration of binding media determination by Edman degradation. We determined the amino acid sequence of3animal glues and obtained their differences. But we can’t obtain the expected results of sample’s determination.
