Objective To observe the level changes of tumor necrosis factor-a (TNF-a)and phosphorylated activation transcription factor-2(p-ATF2) in the Chronic bronchitis rat models’lung tissue with light microscopy by herbal-cake-separated-moxibustion, exploring the mechanism of action, providing experiment basis for the treatment of the chronic bronchitis.Methods This study randomly divided the48Sprague wrote Dawley (SD) rats (male and female half) into A(blank group)、B (model group of CB)、C (herbal-cake-separated-moxibustion group) and D (mild moxibustion group),12in each group.Rats of CB model were maked by modified smoked method. Interferenced by herbal-cake-separated-Moxibustion, and compared with the blank group, model group,and the mild moxibustion group.Draw materials in two times(the before and after treatment),testing the macrophage content in bronchoalveolar lavage fluid of rats in each group and the TNF-α、p-ATF2content in lung tissue, exploring the influence of P38MAPK regulate and control the immune function of Chronic bronchitis rats by herbal-cake-separated-moxibustion.Results (1) The lung tissue pathology change of rats:The herbal-cake-separated-moxibustion and mild moxibustion group have the effect of control the inflammation, and herbal-cake-separated-moxibustion effect is more obvious.(2) Cell count in BALF and AM classification count of rats:Herbal-cake-separated-moxibustion and mild moxibustion can reduce the total number of BALF cells and AM counting of chronic bronchitis rats (P<0.05); The herbal-cake-separated-moxibustion effect is more obvious (P>0.05).(3) P-ATF2expression of lung tissue in rats:Herbal-cake-separated-moxibustion and mild moxibustion can reduce P-ATF2expression of lung tissue in chronic bronchitis rats (P<0.05);The herbal-cake-separated-moxibustion effect is more obvious (P>0.05).(4) TNF-α mRNA and TNF-α protein expression of lung tissue in rats:Herbal-cake-separated-moxibustion and mild moxibustion can reduce TNF-α mRNA and TNF-α protein expression of lung tissue in chronic bronchitis rats (P<0.05);The herbal-cake-separated-moxibustion effect is more obvious(P>0.05).(5) Before the treatment,P-ATF2expression positively correlated with TNF-α mRNA and TNF-α protein expression respectively (r=0.597,r=0.642,P<0.01); After the treatment,P-ATF2expression positively correlated with TNF-α mRNA and TNF-α protein expression respectively (r=0.682,r=0.763,P<0.05)Conclusion (1)Herbal-cake-separated-moxibustion can reduce AM content in BALF of CB rats, and ameliorate inflammation.(2)Herbal- cake-separated-moxibustion can reduce TNF-α and P-ATF2expression in chronic bronchitis rats.And P-ATF2plays an important role in chronic bronchitis inflammation.Herbal-cake-separated-moxibustion can reduce its activation of TNF-α by reducing P-ATF2expression, in order to improve the symptoms of chronic bronchitis.(3) Herbal-cake-separated-moxibustion and mild moxibustion can improve the symptoms of chronic bronchitis rats, but the effect of Herbal-cake-separated-moxibustion is much better.
