Purpose:According to the survey, the world millions of cancer patients, about50%will be different degree of pain, about75%of patients with advanced cancerpain problems, of which40%to50%are of moderate to severe pain, and another25%to30%is very intense pain. Recommended the use of WHO three steps therapycan cause80%to90%of patients with cancer pain can be alleviated, but thereexist some serious problems, such as non narcotic analgesics are oftengastrointestinal irritation and bone marrow inhibition, narcotic analgesics andeasy to cause constipation and other adverse reactions and other defects, therebylimiting the three steps therapy application. In recent years with externaltherapy of traditional Chinese medicine in the treatment of cancer pain has beenmore and more attention, mentor professor Wang Wenping through many years ofclinical practice of Chinese medicine theory basis for development of Zhong yaozhitong paste, for the majority of cancer patients to reduce pain provides arecipe. This topic through the establishment of mouse model of cancer pain, rattail tenderness test, and with three stepped medicine were compared, with Zhongyao zhitong paste and analgesic effect. Through the observation of mouse cancermodels in peripheral blood PGE2(prostaglandin E2) and β-EP (beta-endorphin)change, preliminary discuss Zhong yao zhitong paste in the treatment of cancerpain mechanism for the drug, clinical use and provide a theoretical basis.Material and method:1. material: inbred female BALB/C mice, weighing18-22G, electric thermostatwater bath pot, rat tail tenderness meter (Huaibei China biological),electronic scales, cell counting board, sodium sulfide, mouse beta-endorphin(β-EP) ELISA reagent box, the mouse prostaglandin (PGE2) ELISA reagent box,XS-225A electronic analytical balance, LD4-2A anthos2010centrifuge, automatic quantitative microplate reader. S180tumor cell, morphine hydrochlorideinjection, Zhong yao zhitong paste (homemade), applying ointment, distilledwater.2. Method: through the rat tail tenderness experiment,determination of blankgroup, model group, medication group mouse pain threshold changes in painmedicine is affixed, efficacy analysis. Use the mouse beta endorphin ELISA kit,the mouse prostaglandin ELISA kit, determination of blank group, model group,medication group mouse peripheral blood β-EP and PGE2content, to analysisanalgesic mechanism of Zhong yao zhitong plaster analgesic mechanism.Results:1.Test one:Rat tail tenderness experiment shows that the medicine first day, Zhong yaozhitong plaster group after administration of elevated pain threshold value andthe remaining four set threshold value added more obvious differences, p<0.05with statistical significance. Medication third days, Zhong yao zhitong plastergroup after administration of elevated pain threshold value and the remainingfour set threshold value added more obvious differences, p<0.05withstatistical significance. Medication fifth days, Zhong yao zhitong plaster groupafter administration of elevated pain threshold value and the remaining fourset threshold value added more obvious differences, p<0.05with statisticalsignificance.2.Test twoThe test results show that cancer mouse models groupβ-EP content significantlyincreased compared with healthy controls, p<0.05was statistically significant.Zhong yao zhitong plaster groupβ-EP content compared with the model group wereincreased, p<0.05was statistically significant. Zhong yao zhitong plastertherapy groupβ-EP content and Votalin group morphine group, p<0.05, therewas statistical significance. Zhong yao zhitong plaster therapy groupβ-EPcontent is higher than that of Votalin group, below the morphine group. The test results show that cancer mouse models group PGE2content significantlyincreased compared with healthy controls, p<0.05was statistically significant.Zhong yao zhitong plaster group PGE2content compared with the model group wassignificantly decreased, p<0.05was statistically significant. Zhong yaozhitong paste in the treatment of group PGE2content with Votalin group, morphinegroup, p<0.05, there was statistical significance. Zhong yao zhitong pastein the treatment of group PGE2content below the Votalin group, higher than thatof morphine group.Conclusion:Zhong yao zhitong plaster on cancer mouse model of effective treatment, cansignificantly improve cancer mouse model of pain threshold, the analgesic effectand the three ladder analgesic effect comparison of drug, stronger than Votalin,weak to morphine. Medication first days began to work, and the efficacy ofstability. Zhong yao zhitong plaster on cancer mouse model of analgesic effectmay be related to the stimulation of peripheral blood in β-EP generation andinhibition of PGE2release.
