To Protect The Egg Soup Of Gnrha Super Ovulation Ovarian Ultrastructure Of Rats And The Effect Of Somatic Cell Apoptosis

Objective:The objective of this thesis is to observe the effects of Huluantang on the ovarian ultrastructure and cell apoptosis of ovary in rats under GnRHa controlled ovarian hyperstimulation, and to provide scientific basis of traditional Chinese medicine’s effectiveness for IVF-ET.Methods:40SPF female rats of8weeks were chose as research object, and were randomized into5groups, high-dose medicinal group, moderate-dose medicinal group, low-dose medicinal group,model group, normal group,each group had eight rats.The former four groups were given intraperitoneal Injection of Alarelin,once a day for9days.Till the9th day, HMG was injected,HCG was injected after48hours. From the first day of injection of Alarelinall,medicinal groups were given intragastric administration of high, moderate and low dose Chinese medicinals respectively. Then all rats were executed by neck break in batches after4hours of HCG injection.The following indicators were observed on the day of HCG injection:ovarian ultrastructure by electron microscope; cell apoptosis of ovary through TUNEL.Results:(1)Compared with those in the normal group, the increase of the ovary index in the model group had statistical significance (P＜0.05);Compared with those in the model group, the ovary index of the three medicinal groups were higher than the model group,the increase of the ovary index in the middle dose group had statistical significance(P＜0.01);the increase of the ovary index in the low dose and high dose groups had no statistical significance (P＞0.05).(2)Compared with those in the normal group, the increase of cell apoptosis of ovary in the model group had statistical significance(P＜0.01); Compared with those in the model group, the decrease of cell apoptosis of ovary in all medicinal groups had statistical significance (P＜0.05; P＜0.01).(3)Obverse the morphology of ovary by electron microscope:the cell of ovary in the normal group had more lipid droplets, glycogen and mitochondria;the cell of ovary in the model group shrunk,cytoplasmic vacuoles were swollen,some cells appeared apoptosis state,and the mesenchymal component proliferated obviously;the cell of ovary in the middle dose group had much lipid droplets, some mitochondria were swollen,and the mesenchymal component were more than normal group.Conclusion:1.GnRHa superovulation may have adverse effects to the quality of rats’oocytes.2.The Chinese medicine of Huluantang can increase the ovary index of rats under GnRHa superovulation3.The cell apoptosis of ovary of the rats under GnRHa superovulation increased compare with the normal group before ovulation,and the Chinese medicine of Huluantang can decrease it obviously.4.The Chinese medicine of Huluantang can improve the ovarian ultrastructure of the rats under GnRHa superovulation.
