Objective: Research on Xin Yanning granules in mice caused by CVB3minfection of the viral myocarditis treatment function,andpreliminary discuss the mechanism.Methods:125male BALB/c purebred mice,4to5-week-old,weight16.87±1.13g,randomly divided into5groups: normal control group only20,virus model group only30, treatment to medicine group(high,mediumand low dose group)only25.The mice of the model group and treatmentgroup were infected by0.2ml MEM Eagle liquid of TCID50coxsackievirus B3m(CVB3m) through abdominal cavity injection to induce viralmyocarditis models in Balb/c mice, while normal control group withmethod of the same amount of saline water.The treatment group in0.2ml/10g each time poured the medicine into stomach for14days,andmodel group and normal control group poured the same amount of salinewater into stomach at the same time.The mice of each group had thesame breeding conditions. Dynamic observed general condition of micechanges including weight and mortality rate,and in the3th、5th、7th and14th days from each group took5mice random or remaining mice, observed mice heart quality index (heart quality/weight)、heart visual inspection、the electrocardiogram、myocardial enzyme、light and electron microscopy pathological index.Result:⑴Treatment to medicine group of high dose group of mice’s general conditions were better than model group, and low dose groupwas the similar with model group.In the whole process of observation,total mortality rates were8%(2/25),12%(3/25),20%(5/25),33.3%(10/30).⑵High, medium and lowdose group and model group abnormal ecg rate were13%,36%,40%,55%.Compared with model group,high dose group had obviousdifference(P<0.05),and medium、low dose group had no obviousdifference (P>0.05).⑶Model group in different time myocardialenzyme and pathological integral were increased,comparing withnormal control group had obvious difference(P<0.05). Myocardialenzyme and pathological integral of high and medium dose group inthe5th and7th,14th days were lower than that of the modelgroup,having obvious difference(P<0.01).⑷In the third day,treatment to medicine group and the model of mice heart qualityindexes all had different rate increases;in the5th and7th days,treatment to medicine group and the model of mice heart quality indexes continued to rise,comparing with the model group,high andmedium dose group had significant difference(P<0.01).In the14th day,high and medium dose group of mice heart quality indexes decreased,and low dose group did not have changes, comparing with the modelgroup,high and medium dose group had significant difference(P<0.01).⑸Model group of myocardial ultrastructures change was severer thanthat of treatment to medicine group,and that of low dose group wasalso severer than that of high and medium.Conclusion: Xin Yanning granules can extend the lifetime ofmice,reduce mortality in mice and the incidence of abnormalelectrocardiogram,reduce the myocardial enzyme and heart qualityindex,and reduce the number of animals myocardial pathologicalchanges.
