Swelling And Promote The Spread Of Anal Fistula Postoperative Wound Clinical Research Of Hsp70 Expression

Objective:To observe the influence of getting demibain with xiaozhongcuyusan on expression of heat shock protein70(HSP70) in wound surface tissue of patients after anal fistula operation and also to study its mechanism of effect from the molecular biology level. The evaluation of the clinical curative effect about getting demibain with xiaozhongcuyusan for coalesced wound surface after anal fistula operation was made to provide a true and reliable scientific evidence in the climical application.Methods:Fifty-seven standard anal fistula patients are divided into two groups randomly, which are the therapy group (n=29) and the control group (n=28). Being treated statistically, there is no significant difference between these two groups in age, gender and weight. In other words, these two groups are comparable. The patients of the therapy group receive the demibain two times per day by xiaozhongcuyusan of Mentor’s empirical formula, one in the morning and one in the evening. At first, they are cauterized for5minutes. Then they are prescribed for10minutes in40°C water. The control group are prescribed with liquor warm Nacl as same as the former. The two groups are observed and compared each other after one course of therapy for7days. In the wound surface tissue HSP70expression and content are observed by real-time fluorescence quantitative PCR at the first day of no using medicine and at the third, also the seventh day of using medicine after operation to the two groups, then observing the results with statistical analysis.Results:There all existed gene expression in wound surface tissues after anal fistula operation. There were no difference between the two groups about HSP70,s expression in wound surface tissues at1st postoperative day before the treatment(P>0.05).But in treatment groups, the HSP70 expression quantity were upper to the control group at the3rd and7th postoperative day, there were relatively significant difference between them(P<0.05).Conclusion:Xiaozhongcuyusan has functions of clearing heat and dissolving dampness, of detumescence and acesodyne, convergence and promoting granulation. It can make medicine directly influence the surface of the wound, and improve expression of the wound surface tissue for HSP70. At the same time, the pharmaceuticals have wide resource. The preparation is simple. It is convenient to use and safe in effect. As the result, it is worth making the widely using for clinic.
