| Objective: To observe methicillin-resistant Staphylococcus aureus wound surface sepsis leads to rat acute lung injure and lung tissue's cell apoptosis.Methods and Material: Select eighty Sprague-Dawley(SD) rats weight 250-300g,Male.Randomly divided into two groups.MRSA group and Sodium Chloride group.Each group has 40 ones.Adopt 20 % backside scald modle.Injecting MRSA in scab ,which is called MRSA group, in contrast with the rest ones treated with 0.9% saline solution,which is called control group. Divided rats into five anchor points,at each time point, Observing rat's mentals,appetites,activities,death rates;we cut a piece of tissue from the rat, After tissue homogenate,and dilute,to count the bacterials;Taking rat's lung tissue with HE stain to adopt rank graded;adopt TUNEL method to detect lung cell appoptosis index,watch lung tissue injure condition.Results: All rats in this experimentation were survived. Experiment group rat's general state were unsuperior to control group. No swelling edema and eschar appeared on the wound surface.HE stain shows that after burned for 2,12,24,48h,alveolus spaces are broaden,stroma edema, alveolus cavity are narrow,inflammation cell effusion and little air duct damage,show it's has strong inflammation reaction.TUNEL method found that lung tissue appoptosis index variety with time.In experement group It's arrive to peak when 24 hour while descend at 48 hour.The control group the AI ascend all of the time. The AI of control group are lower than experement group (P<0.05)had significant difference.The rats wound surface tissue had HE stain,we can found tissue putrescence, inflammation reaction is severity,we can judge it was III°burnning.Conclusion MRSA wound surface sepsis rats acute lung injure,the AI is ascending, lung tissue damage aggravates along with the time,the lung tissue cell apoptosis,and the apoptosis index shows that it's ascending in early damage of lung tissue,descending at last. |
