| Objective:To explore the Protective effect of stress response in the surgical trauma. The level of HSP70 in lymphocytes and the level of ALT,TB, Cortisol CRP in serum frome rats which have performed varying degrees of surgical trauma were detected and the relationship between HSP70 and Liver function, GC and CRP was researched.Methods:1. To set up animal models:The 400 healthy male Sprague-Dawley (SD) rats were randomly divided into four groups:(1) control group (GO group):rats were performed without any surgical treatment; (2) experimental groups were:①mild trauma group (G1 group):rats were performed the hepatic left lateral lobectomy;②moderate trauma group (G2 group):rats were performed the hepatic left lateral and right lobectomy;③severe trauma group (G3 group):rats were performed the hepatic left lateral, right and nipple right lobectomy. Each group included.20 rats which were reared in the same environment.2. To detect indicators:Taking blood samples separately in each group at 12h of preoperative,2h,12h,24h and 48h of postoperative. The HSP70 in lymphocytes were detected by using flow cytometry. The ALT,TB and CRP in serum were detected by auto biochemical machine.The cortisol was detected by auto luminescence analyzer. The sata was analyzed by statistical software SPSS17.0.Results:1. Successfully established model of varying degrees surgical trauma by SD rat hepatic lobectomy. The mild traumatic group survival rate was 97.5%; the moderate injury group survival rate was 95.0%; the severe injury group survival rate was 91.3%.The difference between three groups was not significant (P>0.05).2. The HSP70 levels in lymphocytes increased with time after surgical trauma, reaching the peak at 12h (P<0.05), and then decreased slowly until they are higher than the control group level at 48h; The HSP70 levels in lymphocytes increased with varing degrees of surgical trauma (P<0.05),except the moderate trauma group and the severe trauma group at 24h after surgical trauma (P>0.05).3. The ALT levels increased with time after surgical trauma.There were significant differences at 12h after surgical trauma (P<0.05). ALT levels reach the peak at 24h after surgical trauma in three groups (P<0.05), and then gradually declined.At 2h,12h,24h after trauma, the ALT levels increased with varing degrees of surgical trauma (P<0.05). There was no significant difference of TB levels between the mild trauma group and the control group(P>0.05). The TB levels increased with time after surgical trauma in moderate and severe trauma groups.There were significant differences at 12h after surgical trauma (P<0.05). In moderate and severe trauma groups, the TB levels reach the peak at 24h after surgical trauma, then gradually declined.At 12h and 24h after surgical trauma, the TB levels increased with varing degrees of surgical trauma (P<0.05).4. The cortisol levels rapidly reach the peak at 24h after surgical trauma in three groups (P<0.05), and then gradually declined.But the cortisol levels were still higher than control group level in 48h after surgical trauma (P<0.05). At 2h, 12h and 24h after surgical trauma, the cortisol levels increased with varing degrees of surgical trauma (P<0.05).5. The CRP levels kept raising after surgical trauma in three groups,and reach the peak at 24h after surgical trauma. At each time,after surgical trauma, the CRP levels increased with varing degrees of surgical trauma (P<0.05).6. For the relationship between HSP70 levels in lymphocytes and ALT levels in different groups, there were positive correlation in all three groups(P<0.05).There were no significant relationship between HSP70 levels and TB levels in all three groups(P>0.05). For the relationship between HSP70 levels in lymphocytes and the cortisol,CRP,there were highly(r>0.9,P<0.05) positive relationships at almost all the time, except 2h, after surgical trauma with varing degrees of surgical trauma.Conclusion:1. The HSP70 levels in lymphocytes increased with varing degree of trauma.There were relationship between the HSP70 and the liver function. It's prompted that HSP70 plays an important role in inhibiting the liver function to get worse, The HSP70 has a Protective effect of the body.2.There were highly positive relationships between HSP70 and cortisol,CRP at almost all the time after trauma with varing degrees of surgical trauma. It's prompted that there maybe synergy among HSP70 and cortisol,CRP. but the mechanism must be clear by further study. |
